响应面法优化低共熔溶剂提取荔枝果肉多酚及其抗氧化性和酪氨酸酶抑制活性分析

Response surface methodology for optimizing deep eutectic solvent extraction of polyphenols from litchi flesh and analysis of antioxidant and tyrosinase-inhibitory activities

摘要 【目的】开发一种低共熔溶剂提取荔枝果肉多酚的方法,并系统评价其抗氧化性及酪氨酸酶抑制活性,为荔枝果肉的高效利用提供技术支持。【方法】通过低共熔溶剂种类、含水量、料液比、提取温度和提取时间5个单因素及响应面试验优化低共熔溶剂对荔枝果肉多酚的提取条件,评价荔枝果肉多酚的抗氧化性和酪氨酸酶抑制活性,并采用高效液相色谱法(HPLC)分析其主要成分。【结果】确定荔枝果肉多酚的最佳提取条件:摩尔比1∶2的氯化胆碱—乳酸、含水量62.1%、料液比1∶4(g/mL)、提取温度80℃、提取时间60 min,在此条件下荔枝果肉多酚提取量达307.50 mg/100 g。荔枝果肉多酚表现出较强的抗氧化性,对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、2,2'-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐(ABTS)自由基及羟基自由基清除的半抑制浓度(IC_(50))分别为0.051、0.072和0.053 mg/mL,铁离子还原能力为483.50 mg TE/100 g。荔枝果肉多酚对酪氨酸酶抑制活性的IC_(50)(0.031 mg/mL)高于曲酸(0.024 mg/mL),但在0.050~0.160 mg/mL质量浓度范围内,荔枝果肉多酚对酪氨酸酶活性的抑制率高于曲酸。HPLC分析鉴定出芦丁、没食子酸、原儿茶酸、咖啡酸、阿魏酸和对羟基肉桂酸6种主要酚类物质,其中芦丁含量最高(168.97μg/mL)。【结论】本研究优化出一种基于氯化胆碱—乳酸低共熔溶剂的荔枝果肉多酚提取体系,可显著提高荔枝果肉多酚提取量,提取的荔枝果肉多酚具有明显的抗氧化性和酪氨酸酶抑制活性。 【Objective】This study aimed to develop a method for extracting litchi flesh polyphenols(LFP)using a deep eutectic solvent,and systematically evaluate their antioxidant and tyrosinase inhibition activities,with the goal of providing technical support for the efficient utilization of litchi flesh.【Method】Five single factors of deep eutectic solvent(type,water content,material-liquid ratio,temperature,and extraction time)and response surface tests were used to op-timize the extraction conditions of litchi flesh polyphenols with deep eutectic solvent,to evaluate the antioxidant and ty-rosinase inhibition activities of LFP,and to analyze its main components by high performance liquid chromatograph(HPLC).【Result】The optimal extraction conditions were determined:1∶2 molar ratio of choline chloride-lactic acid,wa-ter content of 62.1%,material-liquid ratio of 1∶4(g/mL),extraction temperature of 80°C,time of 60 min,under which the yield of LFP reached 307.50 mg/100 g.LFP exhibited a strong antioxidant activity:its half-maximal inhibitory concen-tration(IC_(50))value was 0.051 mg/mL for scavenging 1,1-diphenyl-2-picrylhydrazyl(DPPH)radicals;its IC50value was 0.072 mg/mL for scavenging 2,2'-azino-bis(3-etilbenzotiazolin)-6-sulfonic acid(ABTS)radicals;its IC_(50)value was 0.053 mg/mL for scavenging hydroxyl radicals;and the ferric reducing ability of plasma was 483.50 mg TE/100 g.The IC50 of litchi flesh polyphenols for tyrosinase inhibitory activity(0.031 mg/mL)was higher than that of kojic acid(0.024 mg/mL);however,the inhibition of tyrosinase by LFP was higher than that of kojic acid in the mass concentration range from 0.050 to 0.160 mg/mL.HPLC analysis identified six major phenolics(rutin,gallic acid,protocatechuic acid,caffeic acid,ferulic acid,p-hydroxycinnamic acid),and the content of rutin was the highest(168.97μg/mL).【Conclusion】This study optimizes a extraction system for litchi flesh polyphenols based on the deep eutectic solvent choline chloride-lactic acid,and it can significantly enhance the yield of litchi flesh polyphenols and the extracted polyphenols in litchi flesh exhibit pronounced antioxidant and tyrosinase inhibition activities.
