八角内生黑曲霉BGEF25产纤维素酶的发酵条件及其酶学性质

Fermentation conditions and enzymatic properties of cellulase produced by endophytic Aspergillus niger BGEF25 of Illicium verum

摘要 试验旨在探究八角内生真菌黑曲霉(Aspergillus niger)BGEF25产纤维素酶的发酵条件及其酶学性质。试验采用单因素法,以羧甲基纤维素酶(CMCase)活性为指标,探讨黑曲霉BGEF25产CMCase的发酵条件。结果显示,以60 g/L的稻草粉作为碳源、15 g/L黄豆粉作为氮源、发酵培养基初始pH值6.0、发酵温度30℃、接种量10%、装液量100 mL(250 mL三角瓶)、摇床转速140 r/min、发酵时间96 h时,CMCase活性最高,可达205 U/mL。酶学性质研究结果显示:该菌株所产纤维素酶系中的CMCase、滤纸酶、β-葡萄糖苷酶的最适pH值分别为5.0、5.0、4.5,最适温度分别为45、50、50℃;在pH值4.0~6.0、温度40~60℃的条件下,3种酶表现出较好的稳定性。MnCl_(2)和ZnSO_(4)等金属盐可提升酶活性,而HgCl_(2)和AgNO_(3)则显著抑制酶活性。研究表明,八角内生真菌黑曲霉BGEF25具有优良的产纤维素酶能力。 The experiment aimed to study the fermentation conditions and enzymatic properties of cellulase produced by endophytic Aspergillus niger BGEF25 of Illicium verum.Using the single-factor method with carboxymethyl cellulase(CMCase)activity as the indicator,the fermentation conditions for CMCase production by Aspergillus niger BGEF25 were investigated.The results showed that when using 60 g/L rice straw powder as the carbon source,15 g/L soybean powder as the nitrogen source,with an initial pH value of 6.0 for the fermentation medium,a fermentation temperature of 30℃,a inoculation rate of 10%,a liquid volume of 100 mL(250 mL test tube),a shaking speed of 140 r/min,and a fermentation time of 96 hours,the activity of the CMCase enzyme was the highest,reaching 205 U/mL.Enzymatic characterization showed that the optimal pH values for CMCase,filter paper enzyme,andβ-glucosidase in the cellulase system were 5.0,5.0,and 4.5,respectively,while their optimal temperatures were 45,50,and 50℃,respectively.All three enzymes exhibited good stability within pH 4.0~6.0 and at temperatures of 40~60℃.Metal salts such as MnCl_(2)and ZnSO_(4)can enhance enzyme activity,while HgCl_(2)and AgNO_(3)significantly inhibit enzyme activity.The study indicates that the endophytic Aspergillus niger BGEF25 of Illicium verum has excellent cellulase production capacity.
