原花青素B_(2)抗氧化与抗炎活性及分子机制研究

摘要 [目的]探究原花青素B_(2)(PB2)通过多靶点干预炎症与胰岛素抵抗(IR)的作用机制。[方法]通过体外抗氧化试验评估PB2的糖尿病预防潜力,并利用分子生物学技术揭示PB2对NF-κB炎症信号通路在IR进展中的调控作用。[结果]PB2通过清除自由基显著降低HepG2细胞的ROS水平,并抑制炎症因子IL-6、IL-1β及TNF-α的表达。PB2通过抑制JNK/NF-κB信号级联,阻断过多ROS生成,从而恢复细胞内氧化还原稳态,最终改善胰岛素抵抗。[结论]PB2可通过调控氧化还原平衡与炎症信号通路,调控糖代谢并拮抗炎症反应。 [Objective]To explore the mechanism by which procyanidin B_(2)(PB2)intervenes in inflammation and insulin resistance(IR)through multi-target modulation.[Methods]The potential of PB2 in diabetes prevention was evaluated via in vitro antioxidant assays,and molecular biology techniques were used to reveal the regulatory effect of PB2 on the NF-κB inflammatory signaling pathway during the progression of IR.[Results]PB2 significantly reduced reactive oxygen species(ROS)levels in HepG2 cells by scavenging free radicals and inhibited the expression of inflammatory factors IL-6,IL-1β,and TNF-α.By suppressing the JNK/NF-κB signaling cascade,PB2 blocked excessive ROS production,thereby restoring intracellular redox homeostasis and ultimately improving insulin resistance.[Conclusion]PB2 can regulate glucose metabolism and antagonize inflammatory responses by modulating redox balance and inflammatory signaling pathways.
