滁菊茎叶精油的提取工艺优化、化学成分及抗氧化活性研究 被引量:1

摘要 为优化滁菊茎叶精油提取工艺、分析滁菊茎叶精油的化学成分和抗氧化活性,本研究通过响应面法优化了水蒸气法提取滁菊茎叶精油的工艺,用气相色谱-质谱联用仪(GC-MS)对滁菊茎叶精油成分进行分析,并通过DPPH和ABTS自由基清除实验研究其抗氧化活性。结果表明最优提取工艺条件为:超声处理时间35 min、提取时间2 h、液料比7∶1。在此条件下,滁菊茎叶精油的提取率为0.97%。经GC-MS分析,滁菊茎叶精油共鉴定出86种化合物,其中质量分数最高的化合物是β-芹子烯,其相对质量分数为12.43%;其次是2-茨酮,为12.13%;然后依次是樟脑(11.18%)、新臭根醇(9.73%)、γ-紫穗槐烯(6.80%)、β-桉叶油醇(6.00%)。滁菊茎叶精油对DPPH和ABTS自由基的IC_(50)分别为46.99 mg/mL和2.42 mg/mL,表明滁菊茎叶精油对ABTS自由基的清除能力强于对DPPH的清除能力。 In order to optimize the extraction process and analyze the chemical composition and antioxidant activity of the essential oil from C.morifolium cv.Chuju stems and leaves,response surface methodology(RSM)was performed.The chemical composition of essential oil was analyzed by gas chromatography-mass spectrometry(GC-MS),and the antioxidant activity was investigated through DPPH and ABTS radical scavenging assays.The results indicated that the optimal extraction conditions were determined as ultrasonication treat time of 35 min,extraction time of 2 h,and liquid-to-material ratio of 7∶1.Under these conditions,an extraction yield of the essential oil of 0.97%was achieved.GC-MS analysis identified a total of 86 compounds,with the highest compound beingβ-apigenin(relative content:12.43%),followed by 2-bornanone(12.13%),camphor(11.18%),neo-odonisol(9.73%),γ-amorphene(6.80%),andβ-eudesmsol(6.00%).The IC_(50)values of the essential oil for DPPH and ABTS free radicals exhibited 46.99 mg/mL and 2.42 mg/mL,respectively,indicating that the essential oil has a stronger scavenging ability against ABTS radical than against DPPH radical.
