天祝白牦牛AdipoR 1基因克隆、mRNA组织表达谱及生物信息学分析

摘要 为了研究脂联素受体1(recombinant adiponectin receptor 1,AdipoR 1)介导脂肪因子脂联素在动物骨骼肌和脂肪细胞的作用及对动物生长、脂肪沉积等性状的影响。试验采用RT-qPCR技术分析AdipoR 1在天祝白牦牛不同组织中的相对表达,应用克隆技术获得天祝白牦牛AdipoR 1编码区序列并进行生物信息学分析。结果表明,天祝白牦牛AdipoR 1在肾脏中表达量最高,其次是肌肉和脂肪组织;其CDS全长1127 bp,编码375个氨基酸。生物信息学分析表明,其属于疏水性、非分泌蛋白,亮氨酸含量最高,二、三级结构主要以α-螺旋为主,存在跨膜螺旋。说明本试验获得的天祝白牦牛AdipoR 1基因组织表达及生物信息学分析结果,为牦牛脂肪相关性状的基因调控研究提供理论依据。 In order to study the effect of recombinant adiponectin receptor 1(AdipoR 1)-mediated adipokine adiponectin in animal skeletal muscle and adipocytes and its effect on animal growth,fat deposition and other traits.The relative expression of AdipoR 1 in different tissues of Tianzhu white yak was analyzed by RT-qPCR.The coding region sequence of AdipoR 1 in Tianzhu white yak was obtained by cloning technique and analyzed by bioinformatics.The results showed that the expression level of AdipoR 1 in Tianzhu white yaks was the highest in the kidney,followed by muscle and adipose tissues.Its full length was 1127 bp,encoding 375 amino acids.Bioinformatics analysis indicated that it belonged to a hydrophobic and non-secretory protein,with the highest content of leucine.Its secondary and tertiary structures were mainly composed ofα-helices,and there were transmembrane helices.The results of tissue expression and bioinformatics analysis of the AdipoR 1 gene in Tianzhu white yaks obtained in this study provide a theoretical basis for the research on gene regulation of fat-related traits in yaks.
