摘要
目的研究蓝指海星来源真菌Nectria sp.GXIMD02049的代谢产物及其生物活性。方法采用多种色谱技术分离菌株的次级代谢产物,并利用核磁共振波谱结合文献数据比对方法鉴定代谢产物的结构。运用微孔板法测定代谢产物的抗菌活性,并通过1,1-二苯基-2-三硝基苯肼自由基(1,1-diphenyl-2-picrylhydrazyl radical,DPPH)清除和铁离子还原抗氧化能力(ferric ion reducing antioxidant power,FRAP)评估代谢产物的体外抗氧化活性。结果16个化合物从Nectria sp.GXIMD02049中分离并鉴定为waol A(1)、trans-dihydrowaol A(2)、waol B(3)、nectriapyrone(4)、vermopyrone(5)、foeniculins C(6)、咖啡酸甲酯(7)、反式阿魏酸(8)、邻羟基苯乙酸甲酯(9)、苯乙酸(10)、对羟基苯乙酸甲酯(11)、对羟基苯乙酸(12)、4-羟基苯乙基乙酸酯(13)、对羟基苯乙醇(14)、苯基乙二醇(15)和对羟基苯甲酸(16)。化合物1和3抗藤黄微球菌的MIC值分别为66.10和58.21μmol/L。化合物1抗耐甲氧西林金黄葡萄球菌、表皮葡萄球菌、黏性放线菌、枯草芽孢杆菌、金黄葡萄球菌的MIC值均为264.83μmol/L。化合物3抗耐甲氧西林金黄葡萄球菌、表皮葡萄球菌、黏性放线菌、枯草芽孢杆菌、金黄葡萄球菌的MIC值均为233.21μmol/L。化合物7和8对清除DPPH自由基的IC_(50)值分别为95.25和85.56μmol/L,它们在50μg/mL下能够还原铁离子,FRAP值分别为406.54和481.85μmol/L。其余化合物在测试浓度下未显示抑菌活性和抗氧化活性。结论化合物1~3、5和6首次从丛赤属Nectria分离。本文丰富了丛赤属Nectria次级代谢产物的多样性。海星共附生真菌Nectria sp.GXIMD02049能够产生抗菌和抗氧化活性物质。
Objective To investigate secondary metabolites of the fungus Nectria sp.GXIMD02049 derived from Linxkia laevigata and their biological activities.Methods The secondary metabolites were separated using the various chromatographic techniques.The structures of the metabolites were identified by NMR spectroscopy and compared with the literature data.The antibacterial activity of the metabolites was determined by the microplate method.The in vitro antioxidant activity of metabolites was evaluated by DPPH free radical scavenging assay and iron ion reduction assay.Results Sixteen compounds were isolated and identified as waol A(1),trans-dihydrowaol A(2),waol B(3),nectriapyrone(4),vermopyrone(5),foeniculin C(6),methyl caffeate(7),trans-ferulic acid(8),methyl O-hydroxyphenylacetate(9),phenyl acetic acid(10),methyl p-hydroxyphenylacetate(11),p-hydroxyphenylacetic acid(12),4-hydroxy phenethyl acetate(13),p-hydroxyphenethyl alcohol(14),1-phenyl-1,2-ethanediol(15),and p-hydroxybenzoic acid(16).Compounds 1 and 3 showed antibacterial activity against Micrococcus luteus with MIC values of 66.10 and 58.21μmol/L,respectively.With an MIC value of 264.83μmol/L,compound 1 showed antibacterial activity against methicillin-resistant Staphylococcus aureus,Staphylococcus epidermidis,Actinomyces viscosus,Bacillus subtilis,and Staphylococcus aureus,and compound 3 had an MIC value of 233.21μmol/L.Compounds 7 and 8 exhibited the activity of scavenging DPPH free radicals with IC_(50) values of 95.25 and 85.56μmol/mL,respectively.Compounds 7 and 8 showed the activity of iron reduction with FRAP values of 406.54 and 481.85μmol/L,respectively,at the concentration of 50μg/mL.Other compounds showed no antibacterial or antioxidant activities at the tested concentration.Conclusion Compounds 1-3,5 and 6 were isolated from Nectria for the first time.This paper expanded the diversity of secondary metabolites of Nectria.The starfish-associated fungus Nectria sp.GXIMD02049 can produce secondary metabolites with antibacterial and antioxidant bioactivity.
