SD大鼠尾部冻伤模型制备及病理分析

Preparation and pathological analysis of a tail frostbite model in SD rats

摘要 目的:探究SD大鼠尾部冻伤模型的制备条件,建立轻度冻伤模型,用于后续新型冻伤防治凝胶对冻伤防治疗效的探究。方法:采用酒精浸泡法制备SD大鼠尾部冻伤模型,实验组大鼠尾部分别在-25℃90%乙醇中浸泡1、3、5 min,在冻伤操作后24 h时取冻伤组织标本,进行常规HE染色,通过冻伤部位表观指标及病理指标对建立的冻伤模型分度。结果:空白对照组大鼠尾部皮肤颜色正常,无充血水肿、无水疱,皮肤完整;1 min组大鼠在冻伤操作24 h后,尾部皮肤无充血水肿、无水疱形成,病理结果显示伤及表皮层;3 min组大鼠冻伤24 h后,尾部皮肤呈红紫色,有充血水肿,出现水疱,病理结果显示伤及真皮层,皮下组织区域血管淤血,部分大鼠出现血管损伤;5 min组大鼠冻伤24 h后,尾部皮肤呈蓝紫色,充血水肿明显,出现血疱,皮肤破溃,病理结果显示伤及皮肤全层及皮下组织。结论:将大鼠尾部在-25℃90%乙醇中浸泡1 min制备得Ⅱ度冻伤模型,浸泡3 min制备得Ⅱ-Ⅲ度过渡的冻伤模型,浸泡5 min制备得Ⅲ度冻伤模型。 Objective:Investigate the preparation conditions of the SD rat tail frostbite model and establish a mild frostbite model for subsequent investigation of the efficacy of a novel frostbite prevention and treatment gel.Methods:An alcohol immersion method was used to prepare a frostbite model on the tail of an SD rat.The tails of the experimentd group rats were soaked in 90%ethanol at-25℃for 1 min,3 min,5min respectively.A frozen tissue specimen was taken 24 hours after frostbite,and conventional HE staining was performed.The established frostbite model was graded by the apparent and pathological indicators of the frostbite site.Results:In the 1 min group,there was no congestion or edema of the skin,and no blisters formed,and the epidermis was not damaged.In the 3 min group,the skin was reddish-purple,congested and edematous,with the appearance of blisters,and the dermis was damaged.There was severe blood congestion in the subcutaneous tissue area,and some rats showed vascular damage.In the 5 min group,the skin was blue-purple,obviously congested and edematous,with the appearance of blood blisters,and the skin was broken and ulcerated,damaging the subcutaneous tissue.Conclusions:The 1 min group is a model of first degree frostbite,the 3 min group is a model of second-to third degree transitional frostbite,and the 5 min group is a model of third degree frostbite.
