70株猪链球菌的分离鉴定和毒力基因检测及耐药性分析

摘要 为了解广西百色市猪源猪链球菌流行分布、菌株携带毒力基因及对常用抗菌药物耐药情况,采用基质辅助激光解吸电离飞行时间质谱对分离自广西百色市辖12个县(市、区)农村散养的366份健康生猪扁桃体样品的猪链球菌进行鉴定,以聚合酶链反应(PCR)对菌株携带毒力基因进行检测,并用微量肉汤稀释法对菌株开展常用抗菌药物耐药性分析。结果显示,共有70株分离株被鉴定为猪链球菌,样品携菌率为19.1%,检出阳性样品主要以右江河为中轴线聚集分布,并与县域生猪调运流通量呈正相关关系。70株猪链球菌的优势毒力基因型以fbps、gapdh、orf2为主,其中有4株全部携带fbps、gapdh、orf2、mrp、sly、cps2J等6种毒力基因,辖区各县(市、区)分离株毒力基因型分布略有差异。70株分离株对红霉素和克林霉素耐药最高,均为94.3%,其次为四环素耐药(92.3%),对头孢曲松耐药性最低(2.8%);分离株整体耐药情况严重,62株表现为多重耐药,占比88.6%,其中34株为三重耐药,12株四重耐药,11株五重耐药,5株六重耐药;菌株流行的耐药谱型为克林霉素、红霉素和四环素三重耐药。研究结果为临床科学用药对猪链球菌病进行预防和治疗,促进养猪业的健康发展具有参考意义。 In order to know the distribution of Streptococcus suis from pigs in Baise,Guangxi,as well as the virulence genes carried by the strains and their resistance to commonly used antimicrobials,the matrix assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS)was utilized to idenbtify the Streptococcus suis isolated from 366 healthy pigs'tonsil samples,which were acquired from 12 counties(cities,districts)in Baise,Guangxi.The virulence genes carried by the strains were detected by polymerase chain reaction(PCR),and the commonly used antimicrobial resistance analysis of the strains was carried out using micro broth dilution method.The result showed that,a sum of 70 isolated strains were identified as Streptococcus suis,with a sample carrying rate of 19.1%.The positive samples were mainly distributed along the Youjiang River axis and positively correlated with the transportation and circulation volume of pigs in the jurisdiction.The dominant virulence genotypes of the 70 Streptococcus sui s strains were mainly about fbps,gapdh,orf2,and four strains carrying fbps,gapdh,orf2,mrp,sly,cps2J were detected,there was a slight discrepancy in the distribution of virulence genotypes among the strains from different isolated counties(cities,districts).70 Streptococcus suis strains had the highest resistance rate to erythromycin and clindamycin,both of them were 94.3%,followed by tetracycline(92.3%),and the lowest resistance to ceftiaxone(2.8%).The overall resistance of isolated strains was serious,of which 62 strains showed multi-drug resistance,accounting for 88.6%,there were 34 strains being triple resistant,12 strains being fourfold resistant,11 strains being fivefold resistant,and five strains being sixfold resistant among them.The prevalent drug resistance pattern of the strains was triple resistance to clindamycin,erythromycin,and tetracycline.This study would be a reference to select the scientific clinical medication for prevention and treatment of swine streptococosis and promote the healthy development of pig industry.
