摘要
为探究双峰驼血浆外泌体对肝癌细胞增殖和侵袭的作用,采用超速离心法提取双峰驼血浆外泌体,并利用透射电子显微镜和纳米颗粒跟踪分析技术对其形态特征进行观察和鉴定。同时,开展外泌体细胞示踪试验,评估肝癌细胞(HEP3B-luc)对外泌体的摄取情况。之后,将外泌体中表达差异显著的miR-144-5p的模拟物(mimics)和miR-144-5p阴性对照(mimics NC)转染至HEP3B-luc细胞,利用CCK-8法和细胞侵袭试验,检测miR-144-5p对HEP3B-luc细胞增殖和侵袭的影响。结果显示,提取的外泌体呈杯状或圆形,大小分布在50~200 nm之间,HEP3B-luc细胞能够有效摄取外泌体。CCK-8试验结果表明,与对照组相比,mimics组的HEP3B-luc细胞增殖能力显著降低(P<0.05),而mimics NC组无显著差异(P>0.05)。细胞侵袭试验结果显示,mimics组的HEP3B-luc细胞侵袭能力显著低于对照组(P<0.01),而mimics NC组无显著差异。以上结果表明,miR-144-5p能够显著抑制HEP3B-luc细胞的增殖和侵袭能力。
To explore the effects of exosomes in Bactrian camel plasma on the proliferation and invasion of hepatocellular carcinoma(HCC)cells,in this experiment,exosomes in Bactrian camel plasma were extracted by the ultracentrifugation method.Transmission electron microscopy and nanoparticle tracking analysis technology were used to observe and identify their morphological characteristics.Meanwhile,an exosome cell tracing experiment was carried out to evaluate the uptake of exosomes by HCC cells(HEP3B-luc).Subsequently,the mimics of miR-144-5p with significantly differential expression in the exosomes and the miR-144-5p negative control(mimics NC)were transfected into HEP3B-luc cells.The CCK-8 assay and cell invasion assay were employed to detect the effects of miR-144-5p on the proliferation and invasion of HEP3B-luc cells.The results showed that the extracted exosomes were cup-shaped or round,with a size distribution ranging from 50 nm to 200 nm,and HEP3B-luc cells could effectively take up the exosomes.The results of the CCK-8 assay indicated that,compared with the control group,the proliferation ability of HEP3B-luc cells in the mimics group was significantly decreased(P<0.05),while there was no significant difference in the mimics NC group(P>0.05).The results of the cell invasion assay showed that the invasion ability of HEP3B-luc cells in the mimics group was significantly lower than that in the control group(P<0.01),while there was no significant difference in the mimics NC group.The above results suggest that miR-144-5p can significantly inhibit the proliferation and invasion abilities of HEP3B-luc cells.
