鸡源解淀粉芽孢杆菌Y02的分离鉴定及其益生特性研究

摘要 为了探究解淀粉芽孢杆菌的益生特性,通过细菌分离纯化、形态特征、生理生化鉴定和16S rRNA序列分析等从肉鸡盲肠中分离鉴定出1株解淀粉芽孢杆菌Y02,并对其耐酸性、耐盐性、抗菌药物敏感性、抑菌活性、产酶能力、溶血性以及安全性等进行综合评价。结果显示,解淀粉芽孢杆菌Y02能够在pH 4.0、100 mg/mL NaCl正常生长,对环丙沙星、头孢喹肟、头孢曲松、诺氟沙星、庆大霉素等15种抗菌药物敏感,对大肠埃希氏菌、金黄色葡萄球菌和表皮葡萄球菌具有良好的抑制作用,具有产蛋白酶和淀粉酶能力,无溶血性,试验过程中各组肉鸡健康存活、体重增加正常、血液指标和血清生化指标无显著差异。表明解淀粉芽孢杆菌Y02具有良好的安全性,可作为肉鸡养殖生产中潜在的益生菌。 In order to explore the probiotic properties of Bacillus amyloliquefaciens.Bacillus amyloliquefaciens Y02 was isolated and identified from the cecum of broilers by purification,morphological,physiological and biochemical characteristics and 16S rRNA sequence analysis,and the comprehensive evaluations were conducted through acid tolerance,salt tolerance,antimicrobial sensitivity,antibacterial activity,enzyme production capacity,hemolytic activity and safety.The results showed that Bacillus amyloliquefaciens Y02 could normally grow at pH 4.0 and 100 mg/mL NaCl.Y02 was sensitive to 15 antimicrobial,including ciprofloxacin,cefquinome,ceftriaxone,norfloxacin,gentamycin,etc.Y02 inhibited Escherichia coli,Staphylococcus aureus,and Staphylococcus epidermidis,secreted amylase,and protease,and had no hemolysis.During the experimental period,broilers exhibited healthy survival,body weight increased normally,and there were no significant differences in blood routine,and serum biochemical indicators among the groups(P>0.05).In conclusion,Bacillus amyloliquefaciens Y02 had good safety and can be used as a potential probiotic in broilers breeding and production.
