摘要
目的探究甲基莲心碱在体内外对胃癌细胞增殖和转移的作用。方法采用MTT法、5-乙基-2′-脱氧尿苷(EdU)掺入实验、细胞划痕实验和Transwell实验分别检测胃癌细胞的增殖、迁移和侵袭;采用Western blot检测细胞增殖和转移相关蛋白表达。在体内,采用裸鼠皮下注射构建异种移植瘤模型;采用尾静脉注射建立肺转移模型;采用免疫组化法检测增殖相关标志物、转移相关标记物的蛋白水平。结果与甲基莲心碱0μM比较,甲基莲心碱5、10μM治疗显著抑制胃癌细胞的体外增殖、细胞迁移和侵袭力。与甲基莲心碱0μM比较,甲基莲心碱5、10μM治疗显著降低Ki67、MMP2蛋白表达,升高TIMP2蛋白表达。在体内,与对照组比较,甲基莲心碱明显抑制肿瘤生长和肺转移。结论甲基莲心碱具有很强的抗肿瘤活性,是一种很有前途的治疗胃癌的候选药物。
Objective To explore the effects of Neferine on the proliferation and metastasis of gastric cancer cells in vivo and in vitro.Methods MTT assay,5-ethyl-2′-deoxyuridine(EdU)incorporation assay,cell scratch assay and Transwell assay were used to detect the proliferation,migration,and invasion of gastric cancer cells,respectively.Western blot was used to detect the expression of cell proliferation and metastasis related proteins.In vivo,a xenograft tumor model was constructed by subcutaneous injection in nude mice,a lung metastasis model was constructed using tail vein injection.Immunohistochemistry was used to detect the protein levels of proliferation related markers and metastasis related markers.Results Compared with Neferine 0μM group,Neferine 5μM and 10μM treatment significantly inhibited the proliferation,migration,and invasion of gastric cancer cells in vitro.Compared with Neferine 0μM group,Neferine 5μM and 10μM treatment significantly reduced Ki67 and MMP2 protein expression,while increased TIMP2 protein expression.In vivo,compared with the control group,Neferine significantly inhibited the tumor growth and lung metastasis.Conclusion Neferine has strong anti-tumor activity and is a promising candidate drug for the treatment of gastric cancer.
