摘要
目的:研究并提高浙南特色中草药菜头肾的质量标准。方法:描述菜头肾鲜品和干品的外观性状,建立横切面和粉末显微鉴别及以连翘酯苷B为对照的薄层色谱(TLC)鉴别方法;参照2020年版《中国药典》四部通则,对其水分、总灰分、酸不溶性灰分、水溶性浸出物进行测定;采用高效液相色谱法(HPLC)和苯酚硫酸法对其主要成分连翘酯苷B和多糖进行含量测定。结果:菜头肾根的构造属于双子叶植物根的少数情况,具有较大的髓部,粉末显微特征具有爵床科植物特点,TLC方法简便,斑点清晰,专属性强;10批样品测定结果:水分10.4%~12.6%,总灰分3.9%~4.9%,酸不溶性灰分0.05%~0.49%,水溶性浸出物67.1%~81.0%,连翘酯苷B含量0.11%~1.29%,多糖含量24.45%~52.67%;含量测定方法学考察均符合要求。结论:所建立的质量标准方法简便,专属性强、准确度高、重复性好,可用于菜头肾药材的质量控制。初步拟定菜头肾药材质量标准为水分不得过14.0%,总灰分不得过6.0%,水溶性浸出物不得少于60.0%,连翘酯苷B按干燥品计不得少于0.37%,多糖按干燥品计不得少于24.0%。
Objective To study and improve the quality standard for Strobilanthes sarcorrhizus.Methods The appearance and characteristics of S.sarcorrhizus were described.Qualitative identification of S.sarcorrhizus was performed using microscopic identification.Thin-layer chromatography(TLC)was performed for identification with the reference substance of forsythiaside B as the control.According to the general principles of the Chinese Pharmacopoeia(2020 edition,Vol.4),the water content,total ash,acid-insoluble ash and water-soluble extract of S.sarcorrhizus were determined.The content determination method for S.sarcorrhizus was established using high-performance liquid chromatography(HPLC),with forsythiaside B as the quality control index.The content of polysaccharide was determined by phenol-sulfuric acid method using D-glucose as reference substance.Results The structure of the fleshy roots of S.sarcorrhizus belongs to a few cases of dicotyledonous roots,with large pith.The powder microscopic characteristics have the characteristics of jasmine plants.TLC method was specific and simple.The water content,total ash,acid-insoluble ash,water-soluble extractive,the content of forsythiaside B and the content of polysaccharide of all samples varied in the ranges of 10.4%-12.6,3.9%-4.9%,0.05%-0.49%,67.1%-81.0%,0.11%-1.29%and 24.45%-52.67%,respectively,the methodological investigation of content determination meets the requirements.Conclusion The established method can be used for quality control of S.sarcorrhizus.The preliminary quality standard of S.sarcorrhizus are as follows:the water content should not be higher than 14.0%,the total ash should not be higher than 6.0%,the water-soluble extract should not be lower than 60.0%,the content of forsythiaside B should not be lower than 0.37%,and the content of polysaccharide should not be lower than 24.0%calculated based on dried products.
