摘要
骨肉瘤作为儿童及青少年群体高发的原发性恶性骨肿瘤,呈现高度异质性、易转移性及不良预后等特点。PI3K/Akt信号通路作为致癌的关键通路之一,可促进骨肉瘤细胞增殖和迁移、诱导放化疗耐药及抑制其凋亡和自噬等。中医药凭借多靶点协同调控PI3K/Akt信号通路在延缓骨肉瘤发展中展现显著优势。本文初步探讨PI3K/Akt信号通路在骨肉瘤中的分子作用机制,并阐述中医药靶向干预该通路的具体靶点和效果。骨肉瘤中抑制PI3K/Akt信号通路的中药活性成分包括萜类、香豆素类、酚类、生物碱类和其他类化合物,中药复方包括桃红四物汤、复方斑蝥胶囊、补肺汤、抑瘤汤、片仔癀、黄芪-莪术药对。本文综述中医药通过PI3K/Akt信号通路治疗骨肉瘤的研究,为骨肉瘤新药研发和临床应用提供帮助。
Osteosarcoma as a primary malignant bone tumor with high incidence in children and adolescents,presents characteristics of high heterogeneity,easy metastasis,and poor prognosis.PI3K/Akt signaling pathway as one of key pathways in carcinogenesis,can promote proliferation and migration of osteosarcoma cells,induce chemoradiotherapy resistance,and inhibit apoptosis and autophagy.Traditional Chinese medicine and pharmacy demonstrates significant advantages in delaying the development of osteosarcoma by synergistically regulating PI3K/Akt signaling pathway through multiple targets.This article preliminarily explores molecular action mechanism of PI3K/Akt signaling pathway in osteosarcoma,and elaborates on specific targets and effects of targeted intervention of traditional Chinese medicine and pharmacy on this pathway.The active constituent of Chinese materia medica that inhibit PI3K/Akt signaling pathway in osteosarcoma include terpenes,coumarins,phenols,alkaloids,and other compounds,compound prescription of Chinese materia medica include Taohong Siwu Decoction,Compound Cantharidin Capsules,Bufei Decoction,Yiliu Decoction,Pien Tze Huang,couplet medicines of Astragali Radix-Curcumae Rhizoma.This article reviews research on traditional Chinese medicine and pharmacy using PI3K/Akt signaling pathway in the treatment of osteosarcoma,providing assistance for the development of new drugs and clinical application in osteosarcoma.
