赤芝担孢子萌发及其抗氧化活性

Basidiospore Germination and Antioxidant Activity of Ganoderma lingzhi

摘要 以广西野生灵芝菌株TZ53为试材,采用内转录间隔区(ITS)序列的分子生物学分析方法对其进行鉴定,采用单因素试验研究了该菌株的孢子在不同基础培养基配方、温度、pH、保存时间、热刺激和木屑煮出液条件下萌发情况,并通过正交实验进一步优化萌发条件;同时研究了萌发菌丝的抗氧化活性和生长特性,以期为广西赤芝杂交育种提供参考依据。结果表明:该菌株为赤芝(Ganoderma lingzhi)。单因素试验结果显示,其孢子萌发的最佳固体培养基配方是8%琼脂+2%牛肉膏,热刺激时间是40 min,木屑煮出液是梨木,保存时间是1~5 d,最适pH 4.0,温度是25~32℃;正交实验结果显示,最佳萌发因子组合为温度是28℃、保存时间1 d、梨木煮出液20%。孢子萌发的菌丝中的多糖、三萜、黄酮、超氧化物歧化酶活性、羟自由基清除能力和总抗氧化能力有一定的差异,说明其具有一定的抗氧化活性。赤芝孢子初生菌丝菌落形态有4种,不同菌落形态的菌丝长速有差异,其中略蓬松型的菌丝长速最快。 The wild Ganoderma strain TZ53 in Guangxi was used as the test material and was identified by molecular biology analysis of ITS sequence.The spore germination of the strain was studied by single factor test method under different basic medium formula,temperature,pH,storage time,thermal stimulation and sawdust boiled liquid conditions,and the germination conditions were further optimized by orthogonal test.The antioxidant activity and growth characteristics of germinated mycelium were studied,in order to provide reference for hybrid breeding of G.lingzhi in Guangxi.The results showed that the strain TZ53 was Ganoderma lingzhi.The results of single factor test showed that the optimum solid medium was 8%agar+2%beef extract,the optimal heat stimulation time and sawdust boiled liquid,and storage time for the spore germination of G.lingzhi were 40 minutes and pear boiled liquid,for 1-5 days,respectively,under conditions of pH 4.0 at 25-32℃.Further optimization based on orthogonal test result showed that the optimal composition of the culture medium was obtained,the temperature was 28℃,storage time was 1 day,and boiling liquid of Pyrus sorotina sawdust was 20%.There were some differences in polysaccharides,triterpenes,flavonoids,superoxide dismutase activity,hydroxyl radical scavenging ability and total antioxidant capacity during spore germination.It was found that the mycelium of spore germination had certain antioxidant activity.There were four types of colony morphology for the primary hyphae of G.lingzhi,and the growth rate of hyphae varies depending on the colony morphology,and the slightly fluffy type had the fastest growth rate.
