摘要
以家蚕茧为原料来源,先通过高温高压水浴脱胶法提取蚕茧中的丝胶蛋白,并用真空冷冻干燥的方法制备丝胶蛋白粉末;再利用碱性蛋白酶、芽孢杆菌蛋白酶、木瓜蛋白酶、中性蛋白酶、风味蛋白酶分别酶解丝胶蛋白并筛选出制备血管紧张素转换酶(ACE)抑制肽的最佳蛋白酶;最后以丝胶蛋白ACE抑制率为主要指标,水解度、分子质量、肽质量浓度为辅助指标,通过单因素试验优化蛋白酶的丝胶酶解工艺参数。结果表明:最优蛋白酶为碱性蛋白酶,最佳酶解参数为底物质量浓度40 g/L、加酶量7.5%(以底物质量为基准)、酶解温度50℃、酶解pH 8.0、酶解时间20 min,在此条件下酶解产物的ACE抑制率为83.846%、水解度为14.298%、肽质量浓度为6.766 mg/mL,分子质量为11.919 Da。所制得的混合多肽分子质量较低、溶解性好、无色透明且无固体杂质。
Using silkworm cocoons as raw materials,sericin was extracted from silkworm cocoons by high-temperature and high-pressure water bath degumming method,and sericin powder was prepared by vacuum freeze-drying method.Secondly,alkaline protease,spore protease,papain protease,neutral pro-tease,and flavor protease were used to hydrolyze sericin,and the optimal protease for preparing angioten-sin-converting enzyme(ACE)inhibitory peptides was screened.Finally,the ACE inhibition rate of seri-cin was used as the main indicator,and hydrolysis degree,molecular weight,and peptide concentration were used as auxiliary indicators to the optimal sericin hydrolysis process parameters through single factor experiments.The results showed that the optimal protease was alkaline protease,and the best enzymatic hydrolysis parameters were as follows:substrate mass concentration of 40 g/L,enzyme dosage of 7.5%(based on substrate mass),hydrolysis temperature of 50,hydrolysis pH of 8.0,and hydrolysis time of 20 min.Under these conditions,the hydrolysates exhibited an ACE inhibitory rate of 83.846%,hy-drolysis degree of 14.298%,peptide mass concentration of 6.766 mg/mL,and molecular weight of 11.919 Da.The prepared mixed peptides had low molecular weight,good solubility,and were colorless,transparent,and free of solid impurities.
