摘要
目的观察垂黄清脉饮对再狭窄模型兔抗炎、抑制平滑肌细胞异常增殖,防止血栓形成,进而探讨其治疗再狭窄的作用机制。方法选取56只新西兰兔,随机分为空白组、模型组、阿托伐他汀钙组、垂黄清脉饮低、中、高剂量组、中西医结合组;造模完成后,灌胃4周。4周后于兔耳缘静脉取血,通过酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)法检测血清中白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necorsis factor-α,TNF-α)水平,通过免疫组化、蛋白印迹(Western Blotting,WB)、逆转录-聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)法测定兔股动脉蛋白PPARγ、PAI1的表达,采用HE染色观察造模效果及治疗变化。结果对比空白组,模型组兔血清IL-6、TNF-α显著升高(P<0.001),股动脉蛋白过氧化物酶体增殖物激活受体γ(peroxisome proliferator-activated receptorγ,PPARγ)、纤溶酶原激活物抑制剂(plasminogen activator inhibitor-1,PAI1)的表达均差异有统计学意义(P<0.05);对比模型组,随着灌药剂量的增加,垂黄清脉饮低、中、高剂量组IL-6、TNF-α、PAI1成反比,PPARγ呈正比,且垂黄清脉饮中剂量结合阿托伐他汀钙组均变化显著(P<0.001);对比阿托伐他汀钙组,低、中剂量组效果差异无统计学意义,垂黄清脉饮高剂量组和中西药结合组差异有统计学意义(P<0.001);对比中剂量组,中西药结合组差异均有显著统计学意义(P<0.001)。结论垂黄清脉饮能达到抗炎、抑制平滑肌细胞异常增殖,防止血栓形成的目的,从而防治再狭窄的发生,且与阿托伐他汀钙产生协同效应,其作用机制可能与抑制IL-6、TNF-α、PAI1,激活PPARγ相关。
Objective To observe the effect of Chuihuang Qingmai Decoction(垂黄清脉饮)on anti-inflammation,inhibition of abnormal proliferation of smooth muscle cells and preventing thrombosis in restenosis model rabbits,and then to explore its mechanism of action in treating restenosis.Methods Fifty-six New Zealand rabbits were randomly divided into blank group,model group,atorvastatin calcium group,Chuihuang Qingmai Decoction low-dose,medium-dose and high-dose groups,and traditional Chinese and western medicine combination group.After the completion of modeling,the rabbits were administered by gavage for 4 weeks.Four weeks later,the blood was collected from the auricular vein of rabbits,and the serum levels of interleukin-6(IL-6)and tumor necorsis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay(ELISA),and the expressions of the femoral artery proteins peroxisome proliferator-activated receptorγ(PPARγ)and plasminogen activator inhibitor-1(PAI1)were determined by immunohistochemistry,Western Blotting(WB)and reverse transcription-polymerase chain reaction(RT-PCR)methods.HE staining was used to observe the effect of model making and the changes of treatment.Results Compared with those in the blank group,the serum levels of IL-6 and TNF-αin the model group were significantly increased(P<0.001),the expressions of femoral artery protein PPARγand PAI1 were different(P<0.05).Compared with those in the model group,with the increase of infusion dose,the levels of IL-6,TNF-αand PAI1 in Chuihuang Qingmai Decoction groups showed inverse ratio,and PPARγshowed positive ratio,and the changes in Chuihuang Qingmai Decoction medium dose combined with atorvastatin calcium group were significant(P<0.001).Compared with that in the atorvastatin calcium group,the effect difference between Chuihuang Qingmai Decoction low dose group and Chuihuang Qingmai Decoction medium dose group was limited,while the difference between Chuihuang Qingmai Decoction high dose group and combination group was significant(P<0.001).Compared with the medium dose group,the combined Chinese and Western medicine group had significant differences(P<0.001).Conclusion Chuihuang Qingmai Decoction can prevent restenosis by anti-inflammation,inhibiting abnormal proliferation of smooth muscle cells and preventing thrombosis,and has a synergistic effect with atorvastatin calcium,the mechanism of which may be related to the inhibition of IL-6,TNF-αand PAI1 and activation of PPARγ.
