摘要 磷脂酶C(phospholipase C,PLC)是细胞信号转导的重要介质。PLC通过产生生物活性分子如肌醇-1,4,5-三磷酸和二酰基甘油调节多种细胞过程。这些产物通过Ca^(2+)的动员、蛋白激酶C和离子通道的激活来调控细胞的增殖、生长和分化。PLCγ1是PLC的主要亚型之一,可直接被膜受体激活,包括受体酪氨酸激酶和黏附受体,如整合素。PLCγ1通过SH结构域与其他信号分子的直接相互作用以及其脂肪酶活性介导信号转导。近年来的研究表明,PLCγ1信号转导的失调会导致机体发育异常,并可能与多种疾病有关。现就PLCγ1的结构、激活机制和生理病理学功能作一综述。 Phospholipase C(PLC)is an important mediator of cellular signal transduction.PLC regulates various cellular processes by generating bioactive molecules such as inositol-1,4,5-triphosphate and diacylglycerol.These products regulate celproliferation,growth,and dfferentiation by mobilizing Ca^(2+),activating protein kinase C and ion channels.PLCγ1 is one of the main subtypes of PLC,which can be directly activated by membrane receptors,including receptor tyrosine kinases and adhesion receptors such as integrins.PLCγ1 directly interacts with other signaling molecules through the SH domain and its lipase activity mediates signal transduction.Recent studies have shown that dysregulation of PLCγ1 signal transduction can lead to developmental abnormalities in the body and may be associated with various diseases.This paper provides a review of the structure,activation mechanism,physiological and pathological functions of PLCγ1.
