摘要
目的探究miR-20b-5p对宫颈癌HeLa细胞增殖、迁移及侵袭能力和凋亡水平的影响。方法利用RT-qPCR比较HeLa细胞和H8细胞miR-20b-5p的表达差异,随后对miR-20b-5p基因分别进行过表达和沉默后,结合CCK-8、迁移实验、基质胶侵袭实验、细胞克隆形成实验以及流式细胞术检测miR-20b-5p对HeLa细胞增殖、迁移和侵袭能力以及细胞凋亡水平。结果He-La细胞miR-20b-5p表达水平高于H8细胞,且其增殖、迁移和侵袭能力均高于H8细胞。miR-20b-5p过表达显著提高了HeLa细胞的增殖、迁移和侵袭能力,降低了其凋亡水平。相反,miR-20b-5p沉默显著降低了HeLa细胞的增殖、迁移和侵袭能力并提高了其凋亡水平。结论miR-20b-5p在HeLa细胞高表达,且与其较高的增殖、迁移和侵袭能力、凋亡水平直接相关,表明其有作为宫颈癌诊断及防控靶点的潜力。
Objective To explore the effect of miR-20b-5p on the proliferation,migration and invasion ability,and apoptosis level of HeLa cells.Methods RT-qPCR was used to compare the expression differences of miR-20b-5p in HeLa cells and H8 cells,and then the miR-20b-5p gene was overexpressed and silenced respectively.CCK-8 assay,migration experiment,matrigel invasion experiment,colony-formation assay and flow cytometry were used to detect the effects of miR-20b-5p on HeLa cell proliferation,migration,invasion ability,and apoptosis levels.Results The expression level of miR-20b-5p in HeLa cells was higher than that in H8 cells,and their proliferation,migration,and invasion abilities were higher than those of H8 cells.Overexpression of miR-20b-5p significantly improved the proliferation,migration,and invasion capabilities of HeLa cells,and reduced their apoptosis levels.On the contrary,silencing of miR-20b-5p significantly reduced the proliferation,migration,and invasion abilities of HeLa cells and increased their apoptosis levels.Conclusions MiR-20b-5p highly expresses in HeLa cells and is directly related to their higher proliferation,migration,invasion capabilities,and apoptosis levels,indicating its potential as a target for the diagnosis and prevention of cervical cancer.
