摘要
本文综述了过去二十年间,本课题组在核苷抗病毒药物发现过程中的工作.综述按照核苷修饰位置进行分类和概述(2′-位和4′-位修饰),以课题组开发的在临床前、临床中和已上市的药物分子为例,主要聚焦在药物研发策略、药物合成、构效关系研究以及临床前和临床研究情况.我们希望本综述能够提高化学、药学和生物学领域工作者对核苷类药物的兴趣,并期望对核苷抗病毒药物的研发起到积极的推动作用.
This review provides the achievements in the discovery of nucleoside antiviral drugs from our group over past two decades.These drug molecules were summarized and classified based on the modification positions of nucleosides,including modifications at 2′-position and 4′-position.This review focused on the drug development strategies,drug synthesis,the structure-activity relationship,as well as pre-clinical and clinical research situations by presenting cases which including drug molecules from our group.We anticipate that the review could not only arouse the interest in nucleoside drugs to a certain degree from the chemical,pharmaceutical and biological communities,but also promote the research and development of nucleoside antiviral drugs.
