瓜蒌润肠汤通过PI3K/AKT信号通路调控小鼠便秘模型的作用机制研究

摘要 目的探究瓜蒌润肠汤治疗便秘的分子作用机制,明确瓜蒌润肠汤治疗便秘的作用效果。方法将40只昆明小鼠分为空白组(灌胃生理盐水)、模型组(灌胃盐酸洛哌丁胺)、阳性组(灌胃盐酸洛哌丁胺+莫沙必利)和中药汤剂治疗组(灌胃盐酸洛哌丁胺+中药汤剂),连续灌胃7 d造模,造模结束后,灌胃阳性药物莫沙必利或中药汤剂7 d,测定粪便含水率、肠道推进率、观察病理切片,并检测血清中炎症因子水平和肠道组织中相关蛋白表达水平。结果相对模型组,灌胃瓜蒌润肠汤可以提高粪便含水率,增加便秘小鼠肠道的推进率,病理切片显示灌胃瓜蒌润肠汤可使肠黏膜表面光滑,腺体排列整齐,杯状细胞增多,此外,血清中VIP和SS含量减少,5-HT和SP水平增高,结肠组织中p-PI3K和p-AKT水平降低,AQP3水平升高。结论瓜蒌润肠汤可以显著改善便秘情况,并通过调节PI3K/AKT信号通路蛋白表达水平调控便秘的发生发展。 Objective To clarify the therapeutic effect of Gualou Runchang Decoction(GRD)on constipation and explore the molecular mechanism of GRD in treating constipation.Methods Fourty Kunming mice were divided into a control group(gavage of physiological saline),a model group(gavage of lopiperidine hydrochloride),a positive group(gavage of lopiperidine hydrochloride+mosapride),and a traditional Chinese medicine decoction treatment group(gavage of lopiperidine hydrochloride+traditional Chinese medicine decoction).Models were made by gavage for 7 consecutive days.After the modeling was completed,the positive drug mosapride or traditional Chinese medicine decoction was gavage for 7 days.Fecal moisture content,intestinal propulsion,pathological sections,serum inflammatory factor levels,and intestinal tissue protein were measured.Results Compared to the model group,oral administration of GRD could increase the fecal moisture content and increase the intestinal propulsion rate of constipated mice.Pathological sections showed that oral administration of GRD could make the surface of the intestinal mucosa smooth,the glandular arrangement neat,increase goblet cells,reduce VIP and SS in serum,increase 5-HT and SP,decrease p-PI3K and p-AKT levels in colon tissue,and increase AQP3 levels.Conclusion GRD can significantly improve constipation and regulate the occurrence and development of constipation by regulating the expression level of PI3K/AKT signaling pathway proteins.
