摘要
目的旨在确认具核梭杆菌(Fusobacterium nucleatum,FN)在结直肠癌(colorectal cancer,CRC)中的作用及相关机制。方法在CRC细胞系中加入FN以检测其对CRC的影响。克隆形成实验检测细胞增殖能力。Transwell法检测细胞侵袭能力。流式细胞技术检测细胞中CD3^(+)CD4^(+)T细胞比例分布情况,Western blotting检测细胞中CD93的表达。与此同时,在细胞中转染si-CD93并后,再采用同上方法分别检测CRC细胞的增殖、侵袭以及CD93蛋白的表达。结果实验结果表明,FN能显著促进CRC细胞的增殖以及侵袭能力(P<0.01)。与此同时,FN能显著上调CD93在CRC细胞中的表达(P<0.01),并能显著提升细胞中CD3^(+)CD4^(+)T细胞的比例(P<0.05)。此外,敲除CD93后,CRC细胞增殖以及侵袭能力减弱(P<0.01),CD93蛋白表达下调(P<0.05)。结论FN能通过上调CRC细胞中CD93的表达,从而促进CRC细胞的恶性程度。
Objective To identify the role and mechanism of Fusobacterium nucleatum(FN)in colorectal cancer(CRC).Methods The effect of FN on CRC was examined by adding it to CRC cell lines.Clone formation assay was used to detect cell proliferation,and Transwell assay was used to detect cell invasion.Flow cytometry was used to detect the distribution of CD3^(+)CD4^(+)T cells and Western blotting was used to detect the expression of CD93 in the cells.At the same time,after transfection of si-CD93 in the cells,the same method was used to detect the proliferation,invasion,and the expression of CD93 protein in CRC cells,respectively.Results The experimental results showed that FN significantly promoted the proliferation and invasion of CRC cells(P<0.01).Meanwhile,FN significantly up-regulated the expression of CD93 in CRC cells(P<0.01)and significantly increased the proportion of CD3^(+)CD4^(+)T cells in the cells(P<0.05).In addition,knockdown of CD93 resulted in diminished proliferation as well as invasion ability of CRC cells(P<0.01),and down-regulation of CD93 protein expression(P<0.05).Conclusion FN can promote malignancy of CRC cells by upregulating CD93 expression in CRC cells.
