摘要
目的观察蜂针治疗,对脑梗死大鼠纤溶系统LPA、tPA/PAI-1的影响,以及脑组织形态学变化、神经功能评分,讨论其抗凝作用及其脑保护作用的机制。方法大鼠适应性喂养1周后,通过线栓法制备大鼠MCAO/R模型,随机分为3组,包括模型组,针刺组、蜂针组,每组6只,共18只。模型组不进行治疗,常规喂养;针刺组采用普通针刺,1~7日每天1针,8~21日每天2针;蜂针组采用穴位活蜂蜇刺,1~7日每天1针,8~21日每天2针。在治疗结束后通过进行Longa评分观察神经功能,TTC染色观察脑梗死面积,采用酶联免疫分析(ELISA)检测大鼠血清LPA、tPA、PAI-1的相关水平。结果蜂针组的大鼠治疗后Longa评分及大脑梗死体积远小于模型组,且差距具有明显的统计学意义(P<0.05);蜂针组和针刺组的大鼠血清中LPA、PAI-1含量均低于模型组,tPA含量均高于模型组,而蜂针组的LPA、PAI-1含量又低于针刺组,tPA含量高于针刺组,且差距具有明显的统计学意义(P<0.05)。结论蜂针疗法可以有效保护脑梗死急性期大鼠神经功能,减少脑梗死面积,具有有效的脑保护作用。其机理可能与减少LPA生成,抑制PAI-1表达,促进tPA表达,从而调节纤溶系统平衡,促使纤维蛋白溶解,抑制血栓生成有关。
Objective To investigate the effects of bee acupuncture on the fibrinolytic system(LPA,tPA/PAI-1),brain tissue morphology,and neurological function scores in rats with cerebral infarction,and to explore the mechanisms underlying its anticoagulant and brain-protective effects.Methods After 1-week of adaptive feeding,the MCAO/R rat model was established by the suture-occluded method.The rats were then randomly divided into 3 groups,with 6 rats in each group,a total of 18 rats.The model group received routine feeding without any treatment.The acupuncture group was treated with ordinary acupuncture(1 needle per day from day 1 to 7;2 needles per day from day 8 to 21),and the bee acupuncture group was subjected to live bee stings at acupoints following the same schedule.After treatment,neurological function was evaluated using the Longa scoring system,the cerebral infarction area was measured by TTC staining,and the serum levels of LPA,tPA,and PAI-1 were determined by ELISA.Results The Longa scores and cerebral infarction volumes in the bee acupuncture group were significantly lower than those in the model group(P<0.05).Both the bee acupuncture and the acupuncture groups had lower serum LPA and PAI-1 levels and higher tPA levels compared to the model group.Moreover,the bee acupuncture group showed more favorable levels than the acupuncture group,with statistically significant differences(P<0.05).Conclusion Bee acupuncture can effectively protect neurological function and reduce the infarction area in rats with acute cerebral infarction.The mechanism may be related to the reduction of LPA production,inhibition of PAI-1 expression,promotion of t-PA expression,thereby regulating the belance of the fibrinolytic system,promoting fibrinolysis,and inhibiting thrombosis.
