摘要
目的研究鱼腥草挥发油对兔耳痤疮模型的治疗效果。方法采用水蒸气蒸馏法制备鱼腥草挥发油,GC-MS法检测鱼腥草挥发油的成分,并通过测定鱼腥草挥发油的最低抑菌浓度评价其抑菌活性;将24只兔随机分为空白对照组、模型组、阳性药物组(维A酸乳膏)及鱼腥草挥发油组;通过涂抹煤焦油14 d,于第7、9、11、13天时在兔耳耳管开口处皮下注射痤疮丙酸杆菌菌液建立兔耳痤疮模型;各组涂抹给药14 d后,肉眼观察兔耳痤疮病变;取兔耳组织制作切片,HE染色观察组织学特征;取兔血清,采用ELISA法检测血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)的水平,评价鱼腥草挥发油对兔耳痤疮的治疗效果。结果在鱼腥草挥发油中鉴定出了126个组分,鱼腥草挥发油对痤疮丙酸杆菌、表皮葡萄球菌、金黄色葡萄球菌的最低抑菌浓度分别为0.281、0.156、0.078 g/L;兔耳痤疮模型给药14 d后,与模型组比较,鱼腥草挥发油组兔耳丘疹、毛囊开口面积有所缩小,兔血清的TNF-α和IL-6水平均有显著降低,差异均具有统计学意义(P<0.05,P<0.01)。结论鱼腥草挥发油具有较好的抑菌活性和抗炎活性,对兔耳痤疮模型有显著的治疗效果。
Objective To study the therapeutic effect of volatile oil from Houttuynia cordata on acne in rabbit ears.Methods The volatile oil of Houttuynia cordata was prepared by using steam distillation,and its components were detected by GC-MS.Its antibacterial activity was evaluated by determining the minimum inhibitory concentration(MIC)of the volatile oil.Twenty-four rabbits were randomly divided into a blank control group,a model group,a positive drug group(tretinoin cream),and a group treated with volatile oil from Houttuynia cordata.By applying coal tar for 14 days,an acne model was established in rabbit ears by subcutaneously injecting Propionibacterium acnes bacterial solution at the opening of the ear canal On the 7 th,9 th,11 th,and 13 th days.After 14 days of medication application in each group,the acne lesions on the rabbit ears were observed visually.Tissue sections were obtained from rabbit ears and stained with HE to observe histological characteristics.Rabbit serum was collected and the levels of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in the serum were detected by using ELISA method to evaluate the therapeutic effect of Houttuynia cordata volatile oil on acne in rabbit ears.Results A total of 126 components were identified in the volatile oil of Houttuynia cordata.The minimum inhibitory concentration(MIC)of Houttuynia cordata volatile oil against Propionibacterium acnes,Staphylococcus epidermidis,and Staphylococcus aureus were 0.281 g/L,0.156 g/L,and 0.078 g/L,respectively.After 14 days of drug administration in the rabbit ear acne model,compared with the modeling group,the group treated with volatile oil from Houttuynia cordata showed a reduction in papules and follicular opening area on the rabbit ears,and the levels of TNF-αand IL-6 in rabbit serum were significantly reduced(P<0.05,P<0.01).Conclusion The volatile oil from Houttuynia cordata exhibits good antibacterial and anti-inflammatory activities,and has significant therapeutic effects on a rabbit ear acne model.
