L-苏氨酸高产菌株的发酵条件优化

Optimization of fermentation conditions for high-yielding L-threonine strains

摘要 目的优化L-苏氨酸高产菌株的发酵条件。方法考察L-异亮氨酸、黄腐酸、甜菜碱盐酸盐、甜菜碱磷酸盐、维生素B3和维生素B4等添加剂对L-苏氨酸发酵的影响,通过响应面实验确定L-苏氨酸高产菌株的最优发酵条件。结果优化后的培养基条件为在发酵培养基中添加200 mg/L L-异亮氨酸和20 mg/L黄腐酸,葡萄糖补料液中添加2.0 g/L甜菜碱盐酸盐、10 mg/L维生素B3、1.5 g/L维生素B4,L-苏氨酸产率达到142.3 g/L,较优化前提高38.1%。结论本研究通过技术创新在L-苏氨酸菌种选育方面实现重大突破,通过对目的菌株发酵培养基的优化,进一步提高L-苏氨酸产量,提高企业经济效益,具有广阔的应用前景。 Objective To optimize the fermentation conditions of the high-yield L-threonine strain.Methods The effects of additives such as L-isoleucine,fulvic acid,glycine hydrochloride,glycine phosphate,vitamin B3 and vitamin B4 on the L-threonine fermentation were investigated.The optimal fermentation conditions of the L-threonine high-yield strain were determined through response surface experiments.Results Under the optimized culture medium conditions,adding 200 mg/L L-isoleucine and 20 mg/L fulvic acid to the fermentation medium,and adding 2.0 g/L glycine hydrochloride,10 mg/L vitamin B3 and 1.5 g/L vitamin B4 to the glucose feed solution,the L-threonine yield reached 142.3 g/L,which was 38.1% higher than that before optimization.Conclusion This study achieves a significant breakthrough in the selection of L-threonine strains through technological innovation.By optimizing the fermentation medium of the target strain,the yield of L-threonine is further increased,improving the economic benefits of the enterprise,and has broad application prospects.
