摘要
目的建立免疫亲和柱前处理结合高效液相色谱法检测薏苡仁、芡实两种经典药食同源物质中黄曲霉毒素B_(1)、B_(2)、G_(1)、G_(2)含量的方法。方法试样用甲醇水提取,经免疫亲和柱净化,以Symmetry■C_(18)柱分离后,经光化学衍生后,采用荧光检测器检测含量。结果黄曲霉毒素B_(1)、G_(1)在0.50~20.00 ng/mL及黄曲霉毒素B_(2)、G_(2)在0.15~6.00 ng/mL范围内线性关系良好,相关系数(r)>0.999。黄曲霉毒素B_(1)、B_(2)、G_(1)、G_(2)的加标回收率在85.07%~95.70%之间,精密度为0.5%~3.4%。结论该方法准确度高且重复性好,为薏苡仁、芡实中黄曲霉毒素B族和G族含量的定性定量分析提供了可靠的技术支持。
Objective To establish a method for the determination of aflatoxin B_(1),B_(2),G_(1) and G_(2) content in 2 kinds of medicinal and food homologous(Coicis semen and Euryales semen)by immunoaffinity column and high performance liquid chromatograph.Methods Samples were extracted with methanol-water,purified through immunoaffinity columns,separated on a Symmetry■C_(18) column,and detected by a fluorescence detector after photochemical derivatization.Results Through methodological validation,aflatoxin B_(1) and G_(1) exhibited good linear relationships within the range of 0.50-20.00 ng/mL,and aflatoxin B_(2) and G_(2) with in 0.15-6.00 ng/mL,with correlation coefficients(r)greater than 0.999.The spiked recoveries for aflatoxin B_(1),B_(2),G_(1) and G_(2) were 85.07%-95.70% with accuracy of 0.5%-3.4%.Conclusion This method demonstrates high accuracy and good repeatability,and can be used for the qualitative and quantitative analysis of aflatoxins B group and G group in Coicis semen and Euryales semen.
