保加利亚乳杆菌与植物乳杆菌发酵槐米药渣中槲皮素的工艺优化

Optimization of process for quercetin in Sophora japonica dregs fermented by Lactobacillus bulgaricus and Lactobacillus plantarum

摘要 试验旨在利用保加利亚乳杆菌和植物乳杆菌发酵槐米药渣,提高槐米药渣中槲皮素的含量。试验在单因素试验的基础上,通过正交试验优化发酵工艺,并采用高效液相色谱法测定发酵前后槐米药渣中槲皮素的含量。结果显示,保加利亚乳杆菌发酵槐米药渣的最佳条件为发酵温度40℃、时间3 d、接种量3 mL,此条件下发酵槐米药渣中的槲皮素含量为0.66181 mg/g。植物乳杆菌发酵槐米药渣的最佳条件为发酵温度30℃、时间5 d、接种量4 mL,此条件下,发酵槐米药渣中的槲皮素含量为0.54994 mg/g。研究表明,试验优化后的发酵工艺稳定有效,能够提高槐米药渣的资源利用率。 The experiment aimed to ferment Sophora japonica dregs using Lactobacillus bulgaricus and Lactobacillus plantarum to increase the content of quercetin in the Sophora japonica dregs.Based on the single-factor experiments,the orthogonal experiments were conducted to optimize the fermentation process.The content of quercetin in the dreg before and after fermentation was determined by high performance liquid chromatography.The results showed that the optimal conditions for Lactobacillus bulgaricus to ferment Sophora japonica dregs were a fermentation temperature of 40℃,a time of 3 days,and an inoculation volume of 3 mL.Under these conditions,the content of quercetin in the fermented dreg was 0.66181 mg/g.The optimal conditions for Lactobacillus plantarum to ferment the Sophora japonica dregs were a fermentation temperature of 30℃,a time of 5 days,and an inoculation volume of 4 mL.Under these conditions,the content of quercetin in the fermented residue was 0.54994 mg/g.The study indicates that the optimized fermentation process was stable and effective,and could improve the resource utilization rate of the Sophora japonica dregs.
