未折叠蛋白反应调控骨稳态平衡机制研究进展

摘要 当细胞受到炎症、基因突变、缺氧等因素的影响,会引起内质网(endoplasmic reticulum,ER)应激。细胞为了恢复ER正常的结构和功能,会启动未折叠蛋白反应(unfolded protein response,UPR)这一适应性调节,目的是为了处理未折叠或折叠错误的蛋白质。如果ER应激状态持续存在,将引起UPR时限的延长,最终诱发细胞凋亡。该文通过总结UPR 3条通路调控成骨细胞与破骨细胞分化的机制研究,重点归纳了蛋白激酶RNA样ER激酶通路在治疗骨质疏松症的重要靶点和机制,探索UPR对于骨稳态平衡机制的影响,旨在为骨质疏松症的治疗提供相关信息。 When cells are affected by factors such as inflammation,gene mutations,hypoxia,etc.,endoplasmic reticulum(ER)stress will be caused.Cells will initiate an adaptive regulation of unfolded protein response(UPR)in order to restore the normal structure and function of the ER,in order to process unfolded or misfolded proteins.If the ER stress state persists,it will cause a prolongation of the UPR time limit,which will eventually induce apoptosis.In this paper,we summarize the mechanism of the three UPR pathways in regulating the differentiation of osteoblasts and osteoclasts,especially the important targets and mechanisms of the protein kinase RNA-like ER kinase pathway in the treatment of osteoporosis,and explore the effect of UPR on the mechanism of bone homeostasis,aiming to provide relevant information for the treatment of osteoporosis.
