摘要
研究以梳唇石斛蒴果为外植体进行组培快繁试验,探究杀菌剂对外植体消毒及生长激素对不定芽、不定根诱导的影响,并筛选出最佳消毒处理、种子萌发培养基、增殖分化培养基、壮苗生根培养基及移栽基质。结果表明:最佳消毒处理为0.08% HgCl2消毒外植体16 min,萌发率达97%;最佳萌发培养基为1/2 MS+蔗糖25 g/L+琼脂5 g/L+马铃薯泥20 g/L,萌发率达98%;最佳增殖分化培养基为MS+蔗糖30 g/L+琼脂5 g/L+马铃薯泥60 g/L+香蕉泥40 g/L+6-BA 0.2 mg/L+NAA 0.4 mg/L,增殖倍数达19.47倍;最佳壮苗生根培养基为3/4 MS+蔗糖20 g/L+琼脂5 g/L+马铃薯泥50 g/L+香蕉泥100 g/L+6-BA 0.5 mg/L+NAA 1.0 mg/L+AC 0.1%,生根率达97.33%,平均生根数5.67条,平均根长5.03 cm;炼苗10 d后,移栽于刨花∶树皮屑=1∶1(体积比)的复合基质中,移栽成活率达95%。
The rapid propagation of Dendrobium strongylanthum was carried out by tissue culture with the capsule as the explant.The effect of disinfectants and growth hormones on the induction of adventitious buds and adventitious roots were studied.Furthermore,the optimal disinfection conditions,seed germination medium,proliferation and differentiation medium,rooting medium,and substrate for transplantation of tissue culture seedlings were screened.The results showed that the optimal disinfection conditions were treatment of the explant with 0.08%HgCl2 for 16 min,under which the germination rate reached 97%.The most suitable medium for seed germination was 1/2 MS+sucrose 25 g/L+agar 5 g/L+potato puree 20 g/L,in which the germination rate was 98%.The optimal proliferation and differentiation medium was MS+sucrose 30 g/L+agar 5 g/L+potato puree 60 g/L+banana puree 40 g/L+6-BA 0.2 mg/L+NAA 0.4 mg/L,in which the proliferation times reached 19.47.The optimal rooting medium was 3/4 MS+sucrose 20 g/L+agar 5 g/L+potato puree 50 g/L+banana puree 100 g/L+6-BA 0.5 mg/L+NAA 1.0 mg/L+AC 0.1%,in which the rooting rate was 97.33%and the average root number and length were 5.67 and 5.03 cm,respectively.After 10 days of acclimatization,the seedlings were transplanted into the compound substrate composed of wood shavings and bark chippings at a volume ratio of 1:1,and the survival rate was 95%.
