超高效液相色谱-电雾式检测器法测定炒玉竹饮片中果糖含量的研究

摘要 目的:建立炒玉竹中果糖含量测定的分析方法。方法:样品用水加热回流提取,采用超高效液相色谱-电雾式检测器法测定,色谱条件:色谱柱为Agilent InfinityLab Poroshell 120 HILIC-Z(2.1 mm×100 mm,2.7μm),流动相为乙腈-0.05%甲酸铵溶液(梯度洗脱),流速为0.45 mL/min,柱温为30℃,进样量为2μL,CAD检测器参数:蒸发温度50℃、幂函数值1.00、数据采集率20 Hz、过滤常数3.6 s,外标法定量。结果:果糖的线性范围为0.1560~2.8079μg(r=0.9999),线性关系良好;重复性及稳定性的RSD均小于2.0%;加样回收率的范围在97.6%~102.7%,RSD为1.9%。10批炒玉竹样品的果糖含量为2.33%~3.67%。结论:所用方法灵敏度、准确度较高、重复性良好,操作简单便捷,可用于炒玉竹中果糖含量的测定。 Objective:To establish an analytical method for determining the content of fructose in Polygonati Odorati Rhizoma Tostum.Methods:The sample was heated and refluxed with water for extraction.UPLC-CAD was used for determination.The chromatographic conditions were as follows:the chromatographic column was Agilent InfinityLab Poroshell 120 HILIC-Z(2.1 mm×100 mm,2.7μm),the mobile phase was acetonitrile-0.05%ammonium formate solution(gradient elution),the flow rate was 0.45 ml min-1,the column temperature was 30℃,the sample volume was 2μL,and the CAD detector detects parameters were as follows:evaporation temperature 50℃,power function value 1.00,data collection rate 20 Hz,filter constant 3.6 s.Quantification was performed using the external standard method.Results:The linear range of fructose was 0.1560~2.8079μg(r=0.9999),with a good linear relationship;The RSD of repeatability and stability are both less than 2.0%;The recovery rate of fructose ranged from 97.6%to 102.7%,with an RSD of 1.9%.The fructose content of 10 batches of Polygonati Odorati Rhizoma Tostum samples was 2.33%~3.67%.Conclusion:The method used has high sensitivity,accuracy,good repeatability,simple and convenient operation,and can be used for the determination of fructose content in Polygonati Odorati Rhizoma Tostum.
