摘要
单细胞转录组测序技术是在单个细胞水平上,通过分离细胞、构建文库、高通量测序及生物信息学分析,获取全基因组表达谱的技术。单细胞转录组测序可获得用于研究特定细胞亚群基因表达异质性的单细胞完整基因表达量数据,有助于识别未知或罕见的细胞类型,并奠定深入研究的基础。本文就单细胞转录组测序方法及其在猪遗传育种、抗病研究、生长发育上的应用进行综述,并探讨其局限性和应用前景。
Single-cell RNA sequencing(scRNA-seq)is a technology that enables the acquisition of whole-genome expression profiles at the individual cell level.This is achieved through a workflow involving cell isolation,library preparation,highthroughput sequencing,and subsequent bioinformatic analysis.scRNA-seq provides comprehensive gene expression data at single-cell resolution,which is instrumental for investigating gene expression heterogeneity within specific cell subpopulations.This capability facilitates the identification of previously unknown or rare cell types and establishes a critical foundation for in-depth mechanistic studies.This review summarizes the methodologies and explores their applications in swine research,with a focus on genetic breeding,disease resistance,and growth and development.Furthermore,the current limitations and future prospects of scRNA-seq technology in these fields are discussed.
