碳酸盐碱度对“鲟龙1号”(Huso dauricus♀×Acipenser schrenckii♂)肝脏代谢的影响

摘要 为探究盐碱胁迫对鲟肝脏的影响,运用LC-MS代谢组学研究经过7 d碳酸盐碱度胁迫后杂交鲟(Huso dauricus♀×Acipenser schrenckii♂)肝脏内源性代谢物的变化,寻找差异代谢物以及相关的代谢通路。结果显示:两个对比组共筛选得到差异显著的代谢物325种,其中135种显著上调,190种显著下调,主要包括L-组氨酸、氧化谷胱甘肽、苯丙氨酸、L-色氨酸、L-缬氨酸、L-苏氨酸、L-异亮氨酸、3-磷酸甘油、1-磷酸半乳糖等;共涉及53条代谢通路,主要有氨基酸生物合成、氨酰-t RNA生物合成、ABC转运蛋白、2-氧代羧酸代谢、甘油酯代谢及糖酵解/糖异生等。推测碳酸盐碱度主要通过影响“鲟龙1号”肝脏氨基酸生物合成、脂质代谢和糖代谢来降低机体中NH3含量,从而避免在高盐碱环境中由于氨排泄受阻导致的氨中毒。本研究可为提高杂交鲟对高盐碱环境的适应能力提供理论依据,对于杂交鲟的养殖和盐碱水的开发利用有重要意义。 In order to explore the effect of carbonate alkalinity stress on liver metabolism of hybrid sturgeon(Huso dauricus♀×Acipenser schrenckii♂),LC-MS metabonomics was used to study the changes of liver metabolites of hybrid sturgeon after carbonate alkalinity stress,and to screen differential metabolites and related metabolic pathways.The result showed compared with the control group,325 significantly different metabolites were screened in the experimental group,of which 135 metabolites were up regulated and 190 metabolites were down regulated.It mainly includes:L-histidinol,oxidized glutathione,L-phenylalanine,L-tryptophan,L-valine,L-threonine,L-isoleucine,glycerol 3-phosphate,galactose 1-phosphate,etc.These differential metabolites involve 53 metabolic pathways,mainly including biosynthesis of amino acids,aminoacyl-tRNA biosynthesis,ABC transporters,2-oxocarboxylic acid metabolism,glycerolipid metabolism,glycolysis/gluconeogenesis,etc.According to the results of this study,it is speculated that in the high saline environment,the hybrid sturgeon liver mainly reduces the content of NH3 in the body through amino acid biosynthesis,lipid metabolism and glucose metabolism,so as to avoid ammonia poisoning caused by blocked ammonia excretion in the high saline environment.This study provides a theoretical basis for improving the adaptability of hybrid sturgeons to high saline alkali environments.This is of great significance for the breeding of hybrid sturgeons and the development and utilization of saline-alkali water.
