基于^(1)H NMR代谢组学的辛酸营养作用机制研究

摘要 中链饱和脂肪酸如辛酸的长期营养作用是人类膳食补充的基础,但研究较少。本研究旨在基于无偏非靶向的核磁共振氢谱法(^(1)H nuclear magnetic resonance spectroscopy,^(1)H NMR)代谢组学,探索辛酸饮食对小鼠的长期营养作用。将40只小鼠随机分为低脂组和辛酸营养组,喂养17月并观察其健康状况。通过^(1)H NMR探索小鼠肝脏内显著差异代谢物和相关代谢途径,系统综合地阐释辛酸饮食营养作用及其机制。结果表明,辛酸可提高雌鼠食物摄入量和存活率,并长期抑制雄鼠肥胖,同时显著降低雄鼠血糖。代谢组学^(1)H NMR研究发现辛酸降低雌雄小鼠肝脏内异亮氨酸、丙氨酸、谷氨酰胺、氧化谷胱甘肽和酪氨酸浓度,升高β-葡萄糖、糖原和苏氨酸水平。此外亮氨酸、α-葡萄糖与腺苷在辛酸组小鼠中表现出性别差异。辛酸营养作用主要影响丙氨酸、天冬氨酸和谷氨酸的代谢;酪氨酸代谢;苯丙氨酸、酪氨酸和色氨酸的生物合成这3条代谢途径。辛酸饮食的营养作用可能通过调控糖酵解、减少氧化应激、降低脂肪沉积、改善胰岛素敏感性和减轻炎症反应等方面预防肝损伤和能量代谢紊乱,从而有助于预防代谢性疾病。 Medium-chain saturated fatty acids,such as octanoic acid,form the basis of human dietary supplements,yet they have been rarely studied.This study aims to explore the long-term nutritional effects of an octanoic acid diet on mice using untargeted proton nuclear magnetic resonance(^(1)H NMR)metabolomics.Forty mice were randomly divided into a low-fat group and an octanoic acid nutritional group,and were fed for 17 months while their health conditions were monitored.The significant differential metabolites in the livers of mice and related metabolic pathways were explored using ^(1)H NMR-based metabolomics to systematically and comprehensively elucidate the nutritional effects and mechanisms of the octanoic acid diet.The results showed that octanoic acid increased food intake and survival rate in female mice,and chronically inhibited obesity in male mice while significantly reducing their blood glucose levels.The ^(1)H NMR-based metabolomics study found that octanoic acid decreased the concentrations of isoleucine,alanine,glutamine,oxidized glutathione,and tyrosine in the livers of both male and female mice,while increasing the levels ofβ-glucose,glycogen,and threonine.In addition,leucine,α-glucose,and adenosine showed gender differences in the octanoic acid group mice.The nutritional effects of octanoic acid mainly affected three metabolic pathways:alanine,aspartate,and glutamate metabolism;tyrosine metabolism;and phenylalanine,tyrosine,and tryptophan biosynthesis.The nutritional effects of octanoic acid diet may prevent liver damage and energy metabolism disorders,thus helping prevent metabolic diseases by regulating glycolysis,reducing oxidative stress,decreasing fat deposition,improving insulin sensitivity,and alleviating inflammatory responses.
