改良Lowry法测定猪口蹄疫O型灭活疫苗总蛋白含量不确定度评定

Measurement Uncertainty Evaluation Determination of Total Protein Content in Swine Foot-and-Mouth Disease Type O Inactivated Vaccine by Modified Lowry Method

摘要 为评定改良Lowry法测定猪口蹄疫O型灭活疫苗总蛋白含量结果的不确定度,分析不确定度的来源和大小,对测定结果作完整表述。识别影响检验结果的准确性主要因素和风险点。针对识别的因素和风险点修订实验室相关体系文件,采取措施消除或控制这些因素和风险点,以达到降低测量不确定度,提升测定结果的准确性和可靠性。采用《口蹄疫灭活疫苗总蛋白含量测定》(农业部公告第2078号)对猪口蹄疫O型灭活疫苗总蛋白含量进行测定。依据《测量不确定度评价与表示》(JJF 1509.1-2012)中相关规定,对改良Lowry法测定过程可能引入的各个不确定度进行分析、评定和量化。得出改良Lowry法测定口蹄疫灭活疫苗总蛋白含量的扩展不确定度U=8.26μg/mL(k=2),发现不确定度主要来源为标准曲线和重复性。 In order to evaluate the uncertainty of total protein content determination in swine Foot-and-mouth disease type O inactivated vaccine using the modified Lowry method,this study analyzed the source and size of the uncertainty,provided a complete description of the determination results.The study identified the main factors and risk points affecting the accuracy of test results.Laboratory-related system documents were revised based on the identified factors and risk points,and steps were taken to eliminate or control these factors and risk points,thereby reducing measurement uncertainty,improving the accuracy and reliability of the measurement results.The total protein content of inactivated Foot-and-mouth disease type O swine vaccine was determined using the"Determination of Total Protein Content of inactivated Foot-and-Mouth Disease Vaccine"(Announcement No.2078,Ministry of Agriculture).Potential uncertainties introduced during the modified Lowry method determination process were analyzed,evaluated,and quantified,in accordance with the relevant provisions of"Evaluation and Expression of Measurement Uncertainty"(JJF 1509.1-2012).The expanded uncertainty was determined as U=8.26μg/mL(k=2)for modified Lowry method in determining total protein content in Foot-and-mouth disease inactivated vaccine.The main sources of uncertainty were identified as the standard curve and repeatability.
