苄星青霉素紫外检测方法的建立及表征

摘要 碘在紫外光区288 nm和354 nm处有最大吸收峰,非离子表面活性剂吐温-20(Tween-20)集脂溶性、水溶性、增敏性于一体,有效改善碘分子的微环境,对碘光吸收有增敏性,苄星青霉素降解产物青霉噻唑酸的还原性改变碘合离子(I_(3)^(-))的氧化状态,引起碘吸光度减小,据此建立苄星青霉素紫外检测新方法。实验结果表明,在波长288 nm处,碘液浓度在1.00×10^(-6)~1.75×10^(-5)mol/L范围内,苄星青霉素浓度与碘的吸光度递减有良好的线性关系,相关系数r=0.9941,检出限为6.41×10^(-7)mol/L。该方法操作简便,可实现苄星青霉素在线、实时、快速微量检测,用于成品药苄星青霉素含量的测定,纯度分析结果在99.91%~101.90%之间。 Iodine has maximum absorption peaks at 288 nm and 354 nm in the ultraviolet region.The non-ionic surfactant Tween-20 combines lipid solubility,water solubility and sensitization,effectively improving the microenvironment of iodine molecules and increasing sensitivity to iodine light absorption.The reducibility of benzathine penicillin degradation product,penicilloic acid,changes the oxidation state of iodide(I_(3)^(-)),causing a decrease in iodine absorbance.Based on this,a new method for UV detection of penicillin was established.The experimental results showed that at a wavelength of 288 nm,there was a good linear relationship between the concentration of benzylpenicillin and the decreasing absorbance of iodine in the range of 1.00×10^(-6 )to 1.75×10^(-5)mol/L of iodine solution,with a correlation coefficient of r=0.9941 and a detection limit of 6.41×10^(-7 )mol/L.This method is easy to operate and can achieve online,real-time,and rapid micro detection of benzylpenicillin.It is used for the determination of benzylpenicillin content in finished drugs,and the purity analysis results are between 99.91%and 101.90%.
