水蛭、烫水蛭中重金属含量及其转移率研究

摘要 采用电感耦合等离子体质谱法测定水蛭和烫水蛭饮片、标准汤剂、配方颗粒中铅(Pb)、镉(Cd)、砷(As)、汞(Hg)元素的含量,并对含量转移率进行评价,为水蛭、烫水蛭配方颗粒的安全性评价提供理论依据。通过聚类分析(CA)确定重金属含量高低与产地的关联性。运用正交偏最小二乘判别分析(OPLS-DA)确定饮片、标准汤剂、配方颗粒的相关性,并筛选影响差异的标志性成分。研究发现,饮片制备成标准汤剂或配方颗粒后,砷元素的含量存在富集的可能性。聚类分析(CA)分析结果显示,部分产自浙江湖州、安徽宿州的水蛭饮片安全性较差。通过正交偏最小二乘判别分析(OPLS-DA),确定了标准汤剂和配方颗粒间较为接近,筛选得到1个VIP值大于1的差异性成分。结果表明,砷元素是影响水蛭、烫水蛭配方颗粒质量安全性的重要因素,饮片中镉、砷元素含量的高低与产地存在一定的关联性,铅元素是影响饮片与标准汤剂或配方颗粒差异性的重要因素。 An inductively coupled plasma mass spectrometry method was carried out for simultaneous determining and evaluating the contents and transfer rates of Plumbum(Pb),Cadmium(Cd),Arsenic(As),Hydrargyrum(Hg)in the decoction pieces,standard decoction and formula granules of Hirudo and its scald product,which could provide a theoretical basis for the safety evaluation of the formula granules of Hirudo and its scald product.The relationship between the contents of heavy metals and the locality of growth was confirmed by cluster analysis(CA).The orthogonal partial least squares discriminant analysis(OPLS-DA)was employed for learning the correlation of decoction pieces,standard decoction and formula granules,and then the signature components which could make discrepancies was screened.The results indicated that the contents of As were might to enrich after the decoction pieces were prepared into standard decoction or formula granules.Some batches of Hirudo decoction pieces,which were produced from Huzhou of Zhejiang and Suzhou of Anhui,were deemed to inferior in the safety quality by CA.The standard decoction and formula granules were close to each other,and a VIP value greater than 1 were screened by OPLS-DA.It was found thar As was the critical factor which could make an impact on the quality safety of formula granules of Hirudo and its scald product.There were a correlation between the contents of Cd and As in the decoction pieces and the place of origin.Pb was an important factor which might cause differences between the decoction pieces and standard decoction or formula granules.
