摘要
为解析新疆伊犁裂腹鱼、塔里木裂腹鱼和新疆高原鳅的生化遗传特性,采用聚丙烯酰胺凝胶电泳技术,分析3种鱼10种组织中的LDH、MDH、EST 3种同工酶。结果表明,3种鱼所有组织中LDH-A基因均占优势(富含A亚基),是其适应新疆高海拔高盐碱环境的结果。伊犁裂腹鱼最多检出3条s-MDH和1条m-MDH酶带;塔里木裂腹鱼最多检出3条s-MDH和3条m-MDH酶带;新疆高原鳅最多检出2条s-MDH和3条m-MDH酶带;酶谱均经典,无基因突变。EST在3种鱼的肝、肾中表达强烈,符合其解毒功能;在伊犁裂腹鱼和塔里木裂腹鱼中表现多态性,在新疆高原鳅中则较单一。该研究揭示了3种鱼的遗传特征,为种质资源保护提供关键依据。
In order to analyze the biochemical and genetic characteristics of Schizothorax pseudaksaiensi,Schizothorax biddulphi and Triplophysa strauchii,the isozymes of LDH,MDH and EST in 10 tissues of three fishes were analyzed by polyacrylamide gel electrophoresis.The results showed that LDH-A gene was dominant in all tissues of the three fishes(rich in A subunit),which was the result of their adaptation to high altitude and high saline-alkali environment in Xinjiang.At most,3 s-MDH and 1 m-MDH bands were detected in S.pseudaksaiensi.A maximum of 3 s-MDH and 3 m-MDH bands were detected in S.biddulphi.A maximum of 2 s-MDH and 3 m-MDH bands were detected in T.strauchii.Enzymogram were classic,no gene mutation.EST was strongly expressed in the liver and kidney of the three kinds of fish,which was consistent with its detoxification function.Polymorphism was found in S.pseudaksaiensi and S.biddulphi,while it was relatively simple in T.strauchii.This study revealed the genetic characteristics of the three fish species and provided a key basis for the protection of germplasm resources.
