摘要
目的考察湿疹喷雾剂的皮肤刺激性、过敏性及对中枢神经系统的影响。方法单次经皮给予家兔湿疹喷雾剂后观察其对完整皮肤及破损皮肤的影响。经皮给予豚鼠湿疹喷雾剂后,观察豚鼠可能出现的过敏反应。通过大鼠自主活动实验、旷场实验、高架十字迷宫实验、协调运动实验及与阈下剂量戊巴比妥钠协同作用实验考察对大鼠中枢神经系统的影响。结果湿疹喷雾剂在120与240μg·ml^(-1)浓度条件下,对兔完整皮肤与破损皮肤无刺激性,豚鼠皮肤未见任何不良反应、致敏率为0。与空白对照组比较,大鼠自主活动、协调运动、探索行为、情绪无差异,湿疹喷雾剂与阈下剂量的戊巴比妥钠无协同作用。结论在本实验剂量水平下,湿疹喷雾剂无皮肤刺激性、过敏性,对大鼠中枢神经系统无影响。
Objective To explore the skin irritation and allergic reactions caused by an eczema spray,and its effects on the central nervous system in rats.Methods The effects of the eczema spray on the intact skin and damaged skin of rabbits were observed after a single transdermal administration.The allergic reactions of guinea pigs after percutaneous administration of the eczema spray were observed.Spontaneous activity experiments,open field experiments,elevated cross maze experiments,coordinated movement experiments,and those on hypnosis of subliminal sleep doses of pentobarbital sodium in rats were conducted to evaluate the effect of the eczema spray.Results At the concentrations of 120 and 240μg:ml^(-1),the eczema spray caused no irritation to the intact or broken skin of rabbits,and no allergic reactions were observed on the skin of guinea pigs.Compared with the control group,there was no difference in spontaneous activity,coordinated movement,exploratory behavior or moods of rats,and the eczema spray had no synergistic effect with sodium pentobarbital at the threshold dose.Conclusion At the dose used in this experiment,the eczema spray induces no skin irritation and allergy reactions,and has no effect on the central nervous system of rats.
