摘要
目的本研究旨在探讨CNGB1在宫颈癌中的表达水平,评估其对患者生存预后的预测价值,并分析其在肿瘤免疫微环境中的潜在作用。方法本研究利用TCGA、GTEx和GEO数据库,通过生物信息学方法分析CNGB1在宫颈癌中的表达及其临床意义。采用Cox回归分析评估CNGB1的预后价值,并构建列线图预测生存期。使用ROC曲线分析CNGB1对宫颈癌的诊断效能。此外,分析CNGB1与免疫检查点的关系,评估其与免疫细胞浸润的相关性,并进行KEGG及GO富集分析探讨CNGB1的潜在功能。结果CNGB1在宫颈癌组织中的表达水平显著高于正常组织(P<0.05)。高CNGB1表达与较差的总生存期(OS)相关,并可作为独立的OS预测因子(P<0.01)。ROC曲线分析显示,CNGB1对宫颈癌的诊断具有较高效能(AUC=0.9362)。在化疗敏感性分析中,CNGB1高表达的宫颈癌患者对多种化疗药物更敏感。免疫分析表明,CNGB1表达水平与CD274、CTLA4、HAVCR2、LAG3、PDCD1等免疫检查点显著相关,并影响宫颈癌组织中免疫细胞的浸润程度。KEGG和GO分析显示,CNGB1主要参与细胞周期、DNA复制及免疫相关信号通路,提示其可能通过影响细胞增殖和肿瘤免疫调控促进宫颈癌进展。结论CNGB1在宫颈癌组织中高表达,并与患者不良预后、化疗敏感性及免疫微环境变化相关。CNGB1可能通过调控细胞周期、DNA复制及免疫微环境促进宫颈癌进展,提示其可作为宫颈癌的预后生物标志物及潜在治疗靶点。
Objective This study aimed to investigate the expression level of CNGB1 in cervical cancer,evaluate its prognostic value for patient survival,and analyze its potential role within the tumor immune microenvironment.Methods Expression profiles and clinical significance of CNGβ1 in cervical cancer were analyzed using bioinformatics approa-ches based on data from the TCGA,GTEx,and GEO databases.Cox regression analyses were performed to assess the prognostic value of CNGβ1,and a nomogram was constructed to predict overall survival(OS).The diagnostic perform-ance of CNGB1 for cervical cancer was evaluated using receiver operating characteristic(ROC)curve analysis.In addi-tion,correlations between CNGB1 expression and immune checkpoints were examined,and its association with immune cell infiltration was assessed.KEGG and GO enrichment analyses were conducted to explore the potential biological functions of CNGβ1.Results CNGβ1 expression was significantly elevated in cervical cancer tissues compared to nor-mal tissues(P<0.05).High CNGB1 expression was associated with poorer overall survival and served as an independ-ent predictor of OS(P<O.01).ROC curve analysis indicated that CNGB1 exhibited high diagnostic efficiency for cervi-cal cancer(AUC=0.9362).Chemotherapy sensitivity analysis revealed that patients with high CNGβ1 expression were more sensitive to multiple chemotherapeutic agents.Immune analysis demonstrated that CNGβ1 expression was signifi-cantly correlated with several immune checkpoints,including CD274,CTLA4,HAVCR2,LAG3,and PDCD1,and influ-enced the degree of immune cell infiltration within cervical cancer tissues.KEGG and GO enrichment analyses suggested that CNGBl was mainly involved in pathways related to the cell cycle,DNA replication,and immune signaling,indicating its potential role in promoting cervical cancer progression by regulating cell proliferation and tumor immune modulation.Conclusion CNGB1 is highly expressed in cervical cancer tissues and is associated with poor prognosis,chemotherapy sensitivity,and alterations in the immune microenvironment.CNGB1 may promote cervical cancer progression by regulating the cell cycle,DNA replication,and immune responses,highlighting its potential as a prognostic biomarker and therapeutic target in cervical cancer.
