摘要
目的筛选健康人群与腹主动脉瘤(AAA)患者外泌体中差异表达的微小RNA(miRNA)。方法2024年1月至2024年8月,收集3例就诊于郑州大学第一附属医院腔内血管外科的男性AAA患者[年龄(63.67±1.53)岁]和3例男性健康对照者(年龄:65.67±2.08岁)的血浆,分离外泌体RNA进行测序。采用miRBase 21数据库识别已知miRNA类型及表达量。使用DEGseq分析两组间显著差异的miRNAs(P<0.05且|log2(差异倍数)|≥1)。通过miRTarBase、miRDB、TargetScan和miRWalk软件预测miRNA靶基因,并基于基因本体论(GO)与京都基因与基因组百科全书(KEGG)富集分析靶基因功能,筛选P<0.05的通路用于可视化。结果AAA组外泌体中共有42种miRNAs表达量显著高于对照组,43种miRNAs表达量显著低于对照组。GO和KEGG富集分析显示这些miRNAs靶基因与多种疾病通路相关。结论AAA患者血浆外泌体miRNA表达谱发生改变,可能通过PI3K-Akt、MAPK、mTOR等信号通路参与AAA的发生发展。
Objective To identify differentially expressed microRNAs(miRNAs)in plasma exosomes between healthy individuals and patients with abdominal aortic aneurysm(AAA).Methods From January 2024 to August 2024,plasma samples were collected from three male abdominal aortic aneurysm(AAA)patients(aged 63.67±1.53 years)who were treated at the Endovascular Surgery Department of the First Affiliated Hospital of Zhengzhou University,along with three age-matched male healthy donors(aged 65.67±2.08 years).Exosomal RNA was subsequently isolated and subjected to RNA sequencing.Known miRNA types and expression levels were identified using the miRBase 21 database.DEGseq was employed to analyze miRNAs with significant differential expression(P<0.05 and|log2(fold change)|≥1).Target genes of miRNAs were predicted using miRTarBase,miRDB,TargetScan,and miRWalk.Functional enrichment analysis of target genes was performed based on gene ontology(GO)and the kyoto encyclopedia of genes and genomes(KEGG),with significantly enriched pathways(P<0.05)selected for visualization.Results A total of 85 significantly differentially expressed miRNAs were identified,including 42 upregulated and 43 downregulated miRNAs.KEGG and GO analyses revealed that the target genes of these miRNAs were associated with multiple disease-related pathways,including PI3K-Akt,MAPK,and mTOR signaling pathways.Conclusion The miRNA expression profile in plasma exosomes of AAA patients is altered and may contribute to AAA pathogenesis through pathways such as PI3K-Akt,MAPK,and mTOR signaling.
