乌腺金丝桃种子休眠特性及破除方法研究

摘要 为筛选促进长白山区乌腺金丝桃种子萌发的最佳条件,以乌腺金丝桃种子为材料,通过测定种子形态、硫酸浸蚀等指标,研究其休眠特性;针对休眠方式,设置不同浓度(400、500、600 mg/L)的赤霉素进行不同时长(6、12、24 h)的浸种处理,并测定种子发芽指标和生理指标。结果表明,硫酸浸蚀乌腺金丝桃种子可显著提高发芽率,说明种皮有透性障碍,存在外源性休眠。另外,种子还含有较高浓度的萌发抑制物,符合种子生理休眠特性。因此,乌腺金丝桃种子的休眠类型为外源性休眠和生理休眠共同存在的综合性休眠;500 mg/L赤霉素浸种处理24 h为最适宜提高其种子发芽效果的方法;赤霉素显著提高了种子中总蛋白、可溶性糖含量及过氧化物酶活性,降低了丙二醛含量。通过对其发芽指标和生理指标隶属函数分析发现,600 mg/L赤霉素浸种处理24 h得分最高。研究表明,600 mg/L赤霉素浸种处理24 h能够最大程度地促进乌腺金丝桃种子的萌发。 To screen out the optimal conditions for promoting the germination of Hypericum attenuatum seeds in the Changbai Mountain area,using Hypericum attenuatum seeds as materials,the dormancy characteristics were studied by measuring seed morphology,sulfuric acid erosion and other indicators.For the dormancy mode,different concentrations of gibberellin(400,500,600 mg/L)were set for seed soaking treatment for different durations(6,12,24 hours),and the germination rate index and physiological indicators of the seeds were determined.The results showed that sulfuric acid erosion of Hypericum attenuatum seeds could significantly increase the germination rate,indicating that there was permeability disorder in the seed coat and exogenous dormancy exists.In addition,the seeds also contained a relatively high concentration of germination inhibitors,which was in line with the physiological dormancy characteristics of seeds.Therefore,the dormancy type of the seeds of Hypericum attenuatum was a comprehensive dormancy where exogenous dormancy and physiological dormancy coexisted.Soaking the seeds in 500 mg/L gibberellin for 24 hours was the most suitable treatment to improve the germination effect of the seeds.Gibberellin significantly increased the total protein,soluble sugar content and peroxidase activity in seeds,while reducing the malondialdehyde content.Through the analysis of the membership functions of its germination index and physiological index,it was found that the score was the highest after soaking seeds in 600 mg/L gibberellin for 24 hours.Study showed that soaking seeds in 600 mg/L gibberellin for 24 hours could promote the germination of Hypericum attenuatum seeds to the greatest extent.
