中国东北地区3种啮齿动物结肠内真菌多样性的比较研究

Comparative study on the diversity of fungi in the colons of three rodent species in Northeast China

摘要 旨在了解啮齿动物肠道真菌的多样性以及鼠类疫源疫病的监测、诊断、治疗和防控。本试验通过高通量测序技术联合培养组学,对东北地区6只黑线姬鼠(Myodes rufocanus)、15只大林姬鼠(Apodemus peninsulae)和18只棕背鼠平(Myodes rufocanus)进行物种相对分布、聚类热图、分类丰度的种间差异、物种分类的Kruskal-Wallis和ANOVA分析。结果:这3种啮齿动物肠道内真菌在门水平上排在前5的种类均包含担子菌门(Basidiomycota)、毛霉门(Mucoromycota)、子囊菌门(Ascomycota)以及捕虫菌门(Zoopagomycota),在属水平上排在前5的均包含丛枝菌根真菌(Rhizophagus),在种水平上3种啮齿动物未发现共有的真菌种类;组间差异分析表明,3种啮齿动物肠道真菌在种水平(R=0.827、P=0.001)和属水平(R=0.777、P=0.001)上均有显著差异,与大林姬鼠和棕背鼠平相比,黑线姬鼠肠道内含有多种特有菌种。综上,本研究初步分离并鉴定了3种啮齿动物的肠道真菌,为进一步研究东北地区啮齿动物肠道真菌菌群差异的成因提供依据。 Rodents are the primary hosts of zoonotic diseases such as plague and encephalitis,and their intestines harbor numerous unknown microorganisms.Among these,fungi play a crucial role as an important component of the intestinal microbiota,contributing to the maintenance of microbial homeostasis and immune regulation.In this study,we utilized high-throughput sequencing combined with culturomics to analyze 39 samples from three rodent species in Northeast China:Myodes rufocanus,Apodemus peninsulae and Myodes rufocanus.The results showed that the top five fungal phyla identified in the intestines of these rodents all included Basidiomycota,Mucoromycota,Ascomycota,and Zoopagomycota.At the genus level,Rhizophagus,a genus of arbuscular mycorrhizal fungi,was among the top five in all species.However,at the species level,significant differences were observed among the three rodents,with no shared fungal species identified.Beta diversity analysis showed that the intestinal fungal communities of the three rodent species differed significantly at both the phylum(R=0.827,P=0.001)and genus(R=0.777,P=0.001)levels.Additionally,the black-striped vole harbored several unique fungal species,compared with the large forest mouse and brown-backed vole.This study initially isolated and identified the intestinal fungi of three rodent species,offering a foundation for further investigation into the factors driving the differences in the intestinal fungal communities of rodents in Northeast China.
