基于代谢组学的不同产地西洋参化学差异研究

摘要 人参皂苷是西洋参主要的次生代谢产物,也是其重要的活性成分。采用超高效液相色谱-高分辨质谱技术对国内外不同产地的西洋参样品进行代谢组学分析,探究其产地差异。基于质量数与电荷的比值m/z、保留时间和二级质谱数据,在不同产地的西洋参样品中定性出74种人参皂苷次生代谢产物。偏最小二乘-判别分析显示国内外不同产地的西洋参样品具有明显分离的趋势。对比美国和加拿大,筛选出22种差异人参皂苷,其在加拿大样品中的含量均显著高于美国样品。对比山东、辽宁和吉林,筛选出12种差异人参皂苷,其中11种在山东产地的含量显著高于辽宁和吉林。最后对比国内和国外样品,筛选出15种差异人参皂苷。以105组差异人参皂苷比值作新变量,筛选出丙二酰化和乙酰化人参皂苷Re的比值变量,作为潜在的化学标志物鉴别国内外样品。丙二酰化/乙酰化Re在国外组和国内组的数值分别为2.92~4.57,4.94~6.65,其验证准确率为83.3%。本研究通过代谢组学分析方法揭示了国内外不同产地西洋参中人参皂苷的差异性,为产地鉴别提供了一种新的策略。 Ginsenosides are the main secondary metabolites and the important active components of Panax quinquefolius.In the study,metabolomics analysis was performed for Panax quinquefolius from different origins at home and abroad based on ultra-high performance liquid chromatography-high resolution mass spectrometry,and the difference among the origins was investigated.Based on molecular weight,retention time and secondary mass spectrometry,74 ginsenosides secondary metabolites were identified from Panax quinquefolius samples of different origins.Partial least squares-discriminant analysis showed that there were significant differences between the domestic and foreign samples.For the comparison of American and Canada,22 differential ginsenosides were screened,and the contents of them from the Canada were significantly higher than those from the American.For the comparison of Shandong,Liaoning and Jilin,12 differential ginsenosides were screened,and the content of 11 differential ginsenosides from the Shandong was significantly higher than that from the Liaoning and Jilin.Finally,for the comparison of domestic and foreign samples,15 differential ginsenosides were selected.105 ratio variables were redefined by the calculation of each two differential ginsenosides,and malonyl/acetyl ginsenoside Re was selected as the potential chemical marker for the identification of samples at home and abroad.The values in the foreign group and the domestic group were 2.92~4.57 and 4.94~6.65,respectively,and the accuracy rate was 83.3%.The study revealed the ginsenosides differences from different origin at home and abroad through the metabolomics analytical method,and proposed a new strategy for the origin distinction.
