Expression and diagnostic value of hsa_circ_16029 in peripheral blood of type 2 diabetes mellitus patients

摘要 目的:通过比较环状RNA16029(hsa_circ_16029)在2型糖尿病(Type 2 Diabetes Mellitus,T2DM)患者组及健康对照(NC)组外周血中表达的差异,评估hsa_circ_16029在T2DM诊断中的价值。方法:(1)收集T2DM组70例及NC组51例外周血样本,通过实时荧光定量聚合酶链式反应(q RT-PCR)外周血hsa_circ_16029的表达水平;(2)相关性分析circ RNA与生化指标之间的相关性;多因素Logistic回归分析circ RNA对T2DM患病风险的影响;受试者工作特征曲线(ROC)评价hsa_circ_16029的诊断价值。结果:(1)q RT-PCR检测结果显示,与NC组相比T2DM组中hsa_circ_16029表达水平显著降低(P<0.001);(2)相关性分析显示,hsa_circ_16029的表达水平与空腹血糖、糖化血红蛋白、HOMA-β存在显著相关性;(3)多因素Logistic回归分析显示,hsa_circ_16029表达水平增高,患T2DM的风险将下降76.3%;(4)ROC曲线显示,hsa_circ_16029的曲线下面积是0.813,特异度为84.3%,灵敏度为68.6%。结论:hsa_circ_16029在T2DM患者外周血中的表达水平显著降低,可能成为T2DM的潜在诊断标志物。 Objective:The value of hsa_circ_16029 in the diagnosis of T2DM was assessed by comparing the differences in the expression of cyclic RNA16029(hsa_circ_16029)in the peripheral blood of a group of patients with type 2 diabetes mellitus(T2DM)and a group of healthy controls(NC).Methods:(1)70 peripheral blood samples from the T2DM group and 51 peripheral blood samples from the NC group were collected,and the expression level of hsa_circ_16029 in peripheral blood of the enrolled subjects was detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).(2)Correlation analysis was performed to analyze the correlation between circRNA and biochemical indexes.Multifactorial Logistic regression analysis was performed to analyze the influence of circRNA on the correlation analysis between circRNA and biochemical indicators.Multifactorial logistic regression analysis of circRNA's influence on the risk of T2DM.Evaluation of the diagnostic value of hsa_circ_16029 by ROC curve.Results:(1)qRT-PCR showed that the expression level of hsa_circ_16029 was significantly lower in the T2DM group compared with the NC group(P<0.001).(2)Correlation analysis showed that the expression level of hsa_circ_16029 had significant correlation with fasting glucose,glycated hemoglobin,and HOMA-β.(3)Multifactorial logistic regression analysis showed that the risk of developing T2DM decreased by 76.3%with higher expression level of hsa_circ_16029.(4)The ROC curve showed that the area under the curve of hsa_circ_16029 was 0.813,the specificity was 84.3%,and the sensitivity was 68.6%(P<0.001).Conclusion:The expression level of hsa_circ_16029 was significantly reduced in the peripheral blood of T2DM patients,which may be a potential diagnostic marker for T2DM.
