温阳督针疗法对缺血缺氧型脑瘫幼鼠干预效果及机制研究

摘要 目的探究温阳督针疗法对缺血缺氧型脑瘫幼鼠的干预效果及潜在机制。方法选择35只7日龄SPF级SD新生乳鼠,10只为假手术组,25只乳鼠制备缺血缺氧脑瘫模型,最终死亡5只,成功造模20只,按照随机数表法将20只脑瘫模型幼鼠分为模型组与针灸组,每组10只。假手术组幼鼠仅分离颈总动脉不结扎,不做缺氧处理。治疗期间,假手术组与模型组仅做与针灸组幼鼠相同的抓取刺激,针灸组采用温阳督针疗法,针刺百会,艾灸大椎和至阳,治疗28 d,治疗结束后苏木精-伊红染色法(HE)染色观察海马组织病理学形态变化,脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(TUNEL)检测海马组织细胞凋亡情况,酶联免疫吸附试验(ELISA)检测血清中白细胞介素(IL)-1β、IL-6、IL-10浓度,蛋白免疫印迹(WB)检测海马组织中磷酸化的酪氨酸激酶-2(p-JAK2)、磷酸化的信号传导子和转录激活子3(p-STAT3)蛋白表达水平。结果模型组幼鼠海马组织出现大量神经细胞损伤固缩、胞质深染,细胞凋亡率显著升高(P<0.05),血清中促炎因子IL-1β、IL-6浓度上升(P<0.05),抗炎因子IL-10浓度下降(P<0.05),海马组织p-JAK2、p-STAT3蛋白表达升高(P<0.05);与模型组比较,针灸组海马组织损伤减轻,凋亡细胞减少,凋亡率降低(P<0.05),血清IL-1β、IL-6浓度下降(P<0.05),IL-10浓度上升(P<0.05),海马组织p-JAK2、p-STAT3蛋白表达降低(P<0.05)。结论温阳督针疗法对缺血缺氧型脑瘫幼鼠具有一定的干预效果,其机制可能与调节炎症因子水平、抑制细胞凋亡、调控JAK2/STAT3信号通路有关。 Objective To explore the intervention effect and potential mechanism of Wenyang Duzhen therapy on ischemic and hypoxic cerebral palsy in young mice.Methods 357-day-old SPF grade SD newborn rats were selected,10 of them were in the sham operation group,and 25 of them were in the hypoxic-ishcemic cerebral palsy model preparation.Finally,5 rats died and 20 rats were successfully modeled.20 young rats with cerebral palsy were divided into the model group and the acupuncture and moxibustion group according to random number table method,with 10 rats in each group.The sham operation group only isolated the common carotid artery without ligation and did not undergo hypoxia treatment.During the treatment,the sham operation group and the model group only used the same grabbing stimulation as the acupuncture and moxibustion group,and the acupuncture and moxibustion group was treated with Wenyang Duzhen therapy,Baihui point was treated with acupuncture,Dazhui point and Zhiyang point were treated with moxibustion for 28 days.After treatment,HE staining was used to observe the pathological changes in hippocampal tissue,TUNEL method was used to detect apoptosis of hippocampal tissue cells,ELISA method was used to detect the levels of interleukin(IL)-1β,IL-6,and IL-10 in serum,and Western blot was used to detect the expression levels of p-JAK2 and p-STAT3 proteins in hippocampal tissue.Results In the model group,a large number of nerve cells in the hippocampal tissue of mice were damaged,shrunk,and deeply stained in cytoplasm;The apoptosis rate increased significantly(P<0.05);And the levels of pro-inflammatory factors of IL-1βand IL-6 in the serum increased(P<0.05),while the level of the anti-inflammatory factor of IL-10 decreased(P<0.05),and the expression levels of p-JAK2 and p-STAT3 proteins in the hippocampal tissue increased(P<0.05).Compared with the model group,the injury of the hippocampal tissue in the acupuncture and moxibustion group was alleviated,the number of apoptotic cells decreased,the apoptosis rate decreased(P<0.05),the levels of IL-1βand IL-6 in the serum decreased(P<0.05),the level of IL-10 increased(P<0.05),and the levels of p-JAK2 and p-STAT3 proteins in the hippocampal tissue decreased(P<0.05).Conclusion Wenyang Duzhen therapy has a certain intervention effect on ischemic and hypoxic cerebral palsy in young mice,and its mechanism may be related to regulating the levels of inflammatory factors,inhibiting cell apoptosis,and regulating the JAK2/STAT3 signaling pathway.
