摘要
目的 建立超高效液相色谱-串联质谱法快速测定黄秋葵果实中20种游离氨基酸的方法。方法 样品经水溶液提取,50%乙腈/水溶液稀释后,采用BEH C_(18)色谱柱分离、0.1%甲酸/水-乙腈溶液为流动相进行洗脱,多反应监测模式测定,外标法定量。结果 20种游离氨基酸在一定质量浓度范围内线性关系良好,相关系数均高于0.996,检出限在0.1~1.5μg/kg之间,3个添加水平下,回收率在79.5%~116.5%之间,相对标准偏差为0.8%~9.8%。结论 该方法可快速准确检测黄秋葵果实中20种游离氨基酸的含量,样品无需干燥处理,检测结果更能准确反映实际成分含量;通过对不同生长期海南主栽黄秋葵果实游离氨基酸含量分析,得出黄秋葵果实成熟期时总游离氨基酸总量、药用氨基酸及必需氨基酸总量等指标呈现最大值,说明成熟期果实具有较高的食疗及药用营养价值,可为海南本地黄秋葵果农采摘期的选择、品质品种筛选和深加工应用提供技术支撑。
Objective To establish a method for rapid determination of 20 kinds of free amino acids in Abelmoschus esculentus(L.)Moench fruits by ultra performance liquid chromatography-tandem mass spectrometry.Methods The samples were extracted with aqueous solution,diluted with 50%acetonitrile-water solution,separated on BEH C18 column,and eluted with mobile phase of 0.1%formic acid/water-acetonitrile solution,determined by multi-reaction monitoring mode,and quantified by external standard method.Results The linear relationships of 20 kinds of free amino acids were good within a certain mass concentration range,with correlation coefficients all above 0.996.The limits of detection ranged from 0.1–1.5μg/kg,under 3 addition levels,the recovery rates were between 79.5%–116.5%,and the relative standard deviations were from 0.8%–9.8%.Conclusion This method can rapid and accurate detection of 20 kinds of free amino acids content in Abelmoschus esculentus(L.)Moench fruits,the samples without the need for drying,and the test results can more accurately reflect the actual component content.Through analysis of free amino acid content in the fruits of the main cultivated Abelmoschus esculentus(L.)Moench in Hainan at different growth stages,it is found that the total amount of free amino acids,medicinal amino acids and essential amino acids reached maximum values during the fruit’s maturity period,this indicates that the fruits at maturity possess higher dietary and medicinal nutritional value.The findings provide technical support for local Hainan Abelmoschus esculentus(L.)Moench farmers in selecting the optimal harvesting period,screening for quality varieties and applications in deep processing.
