39份四川李种质遗传多样性和亲缘关系分析

Analysis of genetic diversity and relationship of 39 plum germplasms in Sichuan Province

摘要 利用ISSR分子标记技术对收集的39份四川李种质资源遗传多样性和亲缘关系进行分析。结果表明,6条引物共扩增出66条条带,平均每条引物可扩增11条条带,多态性条带48条,多态性比率75.74%。供试李种质相似性系数范围为0.59~1.00,平均0.77,等位基因数(Na)和有效等位基因数(Ne)分别为1.970、1.831,Nei’s遗传多样性指数(H)、Shannon’s信息指数(I)和多态信息量(PIC)均值分别为0.443、0.629和0.890,说明供试李种质遗传变幅较大,多样性较丰富。UPGMA聚类结果显示,供试李种质在相似系数0.81处可划分为7个小组(Ⅰ—Ⅶ),组Ⅶ又可进一步划分为4个亚组,划分结果与种质的来源地密切相关。研究结果为四川李种质资源的收集保护和新品种选育提供了重要理论依据。 The genetic diversity and relationship of 39 Prunus salicina L.germplasm resources collected from Sichuan Province were analyzed using ISSR molecular markers.The results showed that 6 primers amplified a total of 66 bands,with an average of 11 bands per primer,including 48 polymorphic bands,yielding a polymorphism rate of 75.74%.The similarity coefficient among the tested plum germplasms ranged from 0.59 to 1.00,with an average of 0.77.The number of alleles(Na)and effective alleles(Ne)was 1.970 and 1.831,respectively.The mean values of Nei's genetic diversity index(H),Shannon's information index(I),and polymorphic information content(PIC)was 0.443,0.629,and 0.890,respectively,indicating a wide genetic variation and rich diversity among the tested plum germplasms.UPGMA clustering results showed that the tested plum germplasms could be divided into 7 groups(Ⅰ—Ⅶ)at a similarity coefficient of 0.81,and groupⅦcould be further divided into 4 subgroups.The classification results were closely related to the origin of the germplasms.The research results provide an important theoretical basis for the collection,protection of plum germplasm resources,and new variety breeding in Sichuan.
