湿热证泄泻大鼠肠道菌群与舌象变化的相关性研究

Correlation study between intestinal flora and tongue image changes in rats with diarrhea of damp-heat syndrome

摘要 目的探究湿热证泄泻大鼠肠道菌群、相关细胞因子及舌象客观化数据三者的相关性。方法“高糖高脂+高温高湿+白酒复合灌胃冰水”构建湿热证泄泻大鼠模型,观察结肠组织形态、舌图象、相关细胞因子表达量及肠道菌群变化,并进行关联性分析。结果模型组大鼠结肠炎性改变明显;血清IL-17A、IL-8、TGF-β、TNF-α、ICAM-1水平,舌质R、S、La值,舌苔S、Lb值,肠道中放线菌门、变形菌门、拟杆菌属、异杆菌属、埃希氏志贺杆菌属、柯林斯氏菌属和肠球菌丰度显著升高,舌质H、I、L值,舌苔I、L值,肠道髌骨菌门、norank_f__Muribaculaceae、梭菌、葡萄球菌属和Candidatus_Saccharimonas丰度显著降低;埃希氏志贺杆菌属和肠球菌与IL-17A、IL-8、舌苔Lb值变化呈正相关,与舌质H、I、L及舌苔L值变化呈负相关,IL-17A、IL-8与舌质H、I、L和舌苔L值变化呈负相关;埃希氏志贺杆菌属还与ICAM-1变化呈正相关。结论血清炎性细胞因子水平和肠道中放线菌门、变形菌门等致病菌丰度上升可能是湿热证泄泻的特征性表现,肠球菌可能与舌质变红相关,埃希氏志贺杆菌属除可能导致舌质变红外,还可能与黄苔、厚腻苔的形成具有相关性。 Objective To explore the correlation among intestinal flora,related cytokines and objective tongue image data in rats with diarrhea of damp-heat syndrome.Methods The rat model of diarrhea with damp-heat syndrome was established busing a“high sugar and fat+high temperature and humidity environment+compound intragastric administration of Baijiu with ice water”.The colonic histopathology,tongue image,expression of related cytokines and changes in intestinal flora were observed,and the correlation analysis was performed.Results The model group exhibited significant colonic inflammatory changes.Serum levels of IL-17A,IL-8,TGF-β,TNF-αand ICAM-1,tongue body R,S and La values,tongue coating S and Lb values,and the abundances of Actinobacteria,Proteobacteria,Bacteroides,Allobaculum,Escherichia-Shigella,Collinsella,and Enterococcus in the intestine were significantly increased.In contrast,tongue body H,I and L values,tongue coating I and L values,and the abundances of Patescibacteria,norank_f__Muribaculaceae,Clostridium,Staphylococcus,and Candidatus_Saccharimonas were significantly reduced.Escherichia-Shigella and Enterococcus showed positive correlations with IL-17A,IL-8,and tongue coating Lb values but negative correlations with tongue body H,I,and L values and tongue coating L values.IL-17A and IL-8 were negatively correlated with tongue body H,I,and L values and tongue coating L values.Escherichia-Shigella was also positively correlated with ICAM-1 levels.Conclusion Elevated serum inflammatory cytokine levels and increased abundances of pathogenic bacteria such as Actinobacteria and Proteobacteria in the intestine may be characteristic manifestations of diarrhea with damp-heat syndrome.Enterococcus might be associated with reddening of the tongue body,while Escherichia-Shigella may contribute not only to tongue redness but also to the formation of yellow and thick greasy tongue coatings.
