基于IL-17信号通路研究益糖康对2型糖尿病糖脂紊乱的影响及作用机制

Study on the effect and mechanism of Yitangkang(益糖康)on glucose and lipid disorders in type 2 diabetes mellitus based on IL-17 signaling pathway

摘要 目的基于IL-17信号通路探讨中药复方益糖康(YTK)对2型糖尿病(T2DM)糖脂代谢紊乱的影响及可能机制。方法将72只SPF级雄性Wistar大鼠,随机分为空白组、模型组、YTK高/中/低剂量组及盐酸二甲双胍组,每组12只,构建T2DM大鼠模型后灌胃给药8周。采用罗氏血糖仪检测空腹血糖(FBG);ELISA法检测空腹胰岛素(FINS),全自动生化分析仪检测甘油三酯(TG)、血清总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C);qRT-PCR和Western blot法检测脂肪组织中IL-17信号通路相关mRNA和蛋白表达水平。结果YTK高、中剂量组大鼠FBG、TG、TC、LDL-C水平显著降低,HDL-C水平明显升高(P<0.05),大鼠脂肪组织IL-17、IL-17R、HSP90、TRAF6、MAPKs、AP-1、IL-6、TNF-α、COX2的mRNA和蛋白表达均显著减少(P<0.05);YTK高、中剂量组IL-17、IL-17R的mRNA表达水平和YTK高剂量组的HSP90、MAPKs、COX2的mRNA表达水平明显低于盐酸二甲双胍组(P<0.05)。结论YTK可以改善T2DM糖脂代谢紊乱,恢复代谢稳态,可能通过抑制IL-17信号通路关键节点与级联反应发挥作用。YTK改善T2DM糖脂代谢紊乱呈量效关系。 Objective To investigate the effect and potential mechanism of the Chinese herbal medicine(CHM)compound Yitangkang(益糖康,YTK)on glucose and lipid metabolism disorders in type 2 diabetes mellitus(T2DM)based on the IL-17 signaling pathway.Methods Seventy-two SPF-grade male Wistar rats were randomly divided into the blank group,model group,YTK high/medium/low-dose groups and metformin hydrochloride group,with 12 in each group.After constructing a T2DM rat model,the patient was gavaged for 8 consecutive weeks.Fasting blood glucose(FBG)was measured using a Roche glucometer;fasting insulin(FINS)was detected by ELISA;triglyceride(TG),total serum cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),and low-density lipoprotein cholesterol(LDL-C)were measured using an automatic biochemical analyzer;qRT-PCR and Western blot(WB)were used to detect the mRNA and protein expression levels of IL-17 signaling pathway-related genes in adipose tissue.Results The levels of FBG,TG,TC,and LDL-C in the high/medium-dose groups of YTK were significantly reduced,and the levels of HDL-C were significantly increased(P<0.05).The mRNA and protein expression of IL-17,IL-17R,HSP90,TRAF6,MAPKs,AP-1,IL-6,TNF-α,and COX2 in the adipose tissue of rats were significantly reduced(P<0.05);the mRNA expression levels of IL-17 and IL-17R in the high/medium-dose groups of YTK and the mRNA expression levels of HSP90,MAPKs,and COX2 in the high/medium-dose groups of YTK were significantly lower than those in the metformin hydrochloride group(P<0.05).Conclusion YTK can alleviate T2DM glycolipid metabolic disorders and restore metabolic homeostasis,which may be associated with inhibiting the reaction between the key nodes of the IL-17 signaling pathway and cascade.Additionally,YTK shows a dosage-efficiency relationship in ameliorating T2DM glycolipid metabolic disorders.
