宫颈癌组织中SHC1、SOS1表达水平与患者病理特征、预后的关系

摘要 目的探讨宫颈癌组织中含有Src同源2结构域的衔接蛋白1(SHC1)、七激酶子同源物1(SOS1)表达水平与患者病理特征、预后的关系。方法选取2016年3月至2022年3月于乐山市市中区妇幼保健院行手术治疗的106例宫颈癌患者作为研究对象,采用免疫组织化学法检测组织中SHC1、SOS1表达水平。比较不同病理特征宫颈癌患者SHC1、SOS1表达差异;术后随访3年,比较不同SHC1、SOS1表达水平患者的预后差异,采用多因素Cox回归分析宫颈癌患者预后的影响因素。结果宫颈癌患者癌组织中SHC1、SOS1阳性率(74.53%、80.19%)高于癌旁组织(33.02%、35.85%)(均P<0.05)。不同分化程度、FIGO分期、淋巴结转移的宫颈癌患者SHC1、SOS1表达差异有统计学意义(P<0.05)。SHC1、SOS1阳性患者3年总生存率为46.84%、49.41%,分别低于SHC1、SOS1阴性患者的88.89%、90.48%(均P<0.05)。多因素分析显示,肿瘤低分化(HR=1.917,95%CI:1.263~2.911)、SHC1阳性(HR=2.765,95%CI:1.645~4.648)、SOS1阳性(HR=3.133,95%CI:1.806~5.434)、淋巴结转移(HR=2.246,95%CI:1.411~3.573)是宫颈癌患者预后的独立危险因素(P<0.05)。结论宫颈癌组织中SHC1、SOS1均呈高表达,其表达水平与宫颈癌患者FIGO分期、肿瘤分化程度、淋巴结转移相关,是影响患者预后的独立危险因素。 Objective To explore the relationship between the expression levels of Src homology 2 domain-containing adaptor protein 1(SHC1)and Son of sevenless homolog 1(SOS1)in cervical cancer tissues and the pathological characteristics and prognosis of patients.Methods A total of 106 patients with cervical cancer who underwent surgery at Shizhong District Maternal and Child Health Hospital from March 2016 to March 2022 were selected.The expression levels of SHC1 and SOS1 in cancer tissues and adjacent tissues were detected by immunohistochemistry.Differences in SHC1 and SOS1 expression among patients with different pathological characteristics were compared.Patients were followed up for 3 years postoperatively,and prognosis differences among those with different SHC1/SOS1 expression levels were compared.Multivariate Cox regression analysis was used to identify factors influencing prognosis.Results The positive rates of SHC1 and SOS1 in cervical cancer tissues(74.53%and 80.19%)were higher than those in adjacent tissues(33.02%and 35.85%)(P<0.05).Significant differences in SHC1 and SOS1 expression were observed among patients with different differentiation degrees,FIGO stages,and lymph node metastasis status(P<0.05).The 3-year overall survival rates of SHC1-positive and SOS1-positive patients were 46.84%and 49.41%,respectively,lower than those of SHC1-negative(88.89%)and SOS1-negative patients(90.48%)(P<0.05).Multivariate analysis showed that poor tumor differentiation(HR=1.917,95%CI:1.263-2.911),SHC1 positivity(HR=2.765,95%CI:1.645-4.648),SOS1 positivity(HR=3.133,95%CI:1.806-5.434),and lymph node metastasis(HR=2.246,95%CI:1.411-3.573)were independent risk factors for prognosis(P<0.05).Conclusion SHC1 and SOS1 are highly expressed in cervical cancer tissues.Their expression levels correlate with FIGO stage,tumor differentiation degree,and lymph node metastasis,serving as independent risk factors for prognosis.
