摘要
目的 明确南京市白蚁种类和系统发育地位。方法 克隆南京市28个白蚁地理种群的线粒体细胞色素氧化酶亚基Ⅱ(COⅡ)基因序列,利用GenBank数据库进行比对分析和分子鉴定,构建系统进化树明确其系统发育关系。结果 基因序列比对结果显示,所测白蚁来自5个属(散白蚁属、乳白蚁属、土白蚁属、近扭白蚁属、楹白蚁属)的12个种,基因序列的A+T碱基含量(63.20%)显著高于G+C含量(36.80%),表现出明显的AT偏好性。遗传距离分析表明,采集的南京市白蚁属间遗传距离为0.160~0.354,种间遗传距离为0~0.354。系统进化树显示散白蚁属和乳白蚁属聚集在一个进化支,其亲缘关系较近、同属于鼻白蚁科;土白蚁属和近扭白蚁属聚为一支,为同属于白蚁科的近缘属;楹白蚁属与其他属亲缘关系较远,单独列为一支。结论 白蚁分子鉴定及系统发育的研究结果与形态学鉴定结论基本一致,为科学鉴定白蚁种类提供了有力证据。
Objective To clarify the termite species and their phylogenetic status in Nanjing.Methods The mitochondrial cytochrome oxidase subunitⅡ(COⅡ)genes of 28 termite populations from different geographical locations in Nanjing were cloned and sequenced.These sequences were aligned using the GenBank database for molecular identification,and a phylogenetic tree was constructed to clarify their phylogenetic relationships.Results Sequence analysis showed that all samples belong to 12 species across 5 genera(Reticulitermes,Coptotermes,Odontotermes,Pericapritermes and Incisitermes).The A+T content of the gene sequences was 63.20%,significantly higher than the G+C content(36.80%),demonstrating a strong AT preference.Genetic distance analysis revealed the inter-generic genetic distances ranged from 0.160 to 0.354,while intra-species distances ranged from 0 to 0.354.Phylogenetic analysis showed that Reticulitermes and Coptotermes clustered together,showing a closer evolutionary relationship within the family Rhinotermitidae,while Odontotermes and Pericapritermes formed a distinct clade within the family Termitidae.In contrast,Incisitermes was more distantly related to the other four genera.Conclusion The results of molecular identification and phylogenetic analysis were basically consistent with traditional morphological identification,providing strong evidence for scientific identification of termite species.
